【摘要】：目的:研究瑞舒伐他汀對(duì)內(nèi)皮細(xì)胞功能變化的影響。方法:選擇2013年6月-2014年6月我院健康體檢者15例,冠心病患者30例。分成健康對(duì)照組、冠心病藥物干預(yù)組、冠心病組,各15例。對(duì)比各組不對(duì)稱二甲基精氨酸(ADMA)、一氧化氮(NO)及一氧化氮合酶(NOS)的血液酶聯(lián)免疫吸附測(cè)定(ELISA)檢測(cè)結(jié)果,分析酶標(biāo)儀光密度(OD)值與其相關(guān)性;對(duì)人臍靜脈內(nèi)皮細(xì)胞(HUVEC)培養(yǎng)4 h及8 h后,根據(jù)瑞舒伐他汀的濃度劑量分成5個(gè)組,即0μmol/L對(duì)照組和0.1、1、10、100μmol/L組,檢測(cè)不同濃度瑞舒伐他汀對(duì)內(nèi)皮細(xì)胞二甲基精氫酸二甲胺水解酶(DDAH)信使核糖核酸(m RNA)的表達(dá)變化。結(jié)果:冠心病藥物干預(yù)組的ADMA水平均顯著低于冠心病組,但仍高于健康對(duì)照組;NO及NOS水平均顯著高于冠心病組,但仍低于健康對(duì)照組,差異均有統(tǒng)計(jì)學(xué)意義(P0.05)。根據(jù)Pearson法分析相關(guān)性可知,酶標(biāo)儀OD值與ADMA、NO及NOS均呈正相關(guān)。HUVEC細(xì)胞培養(yǎng)4 h后的逆轉(zhuǎn)錄-聚合酶鏈反應(yīng)(RT-PCR)結(jié)果顯示,0.1、1、10及100μmol/L瑞舒伐他汀組的DDAH m RNA表達(dá)均上升,且10μmol/L及100μmol/L瑞舒伐他汀組的DDAH m RNA表達(dá)較對(duì)照組的差異有統(tǒng)計(jì)學(xué)意義(P0.05)。培養(yǎng)8 h后的RT-PCR結(jié)果顯示,0.1、1、10及100μmol/L瑞舒伐他汀組的DDAH m RNA表達(dá)均上升,且10μmol/L瑞舒伐他汀組的DDAH m RNA表達(dá)較對(duì)照組的差異有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:瑞舒伐他汀對(duì)內(nèi)皮細(xì)胞具有較好的保護(hù)作用,值得臨床關(guān)注與重視。
[Abstract]:Objective: to study the effect of Risuvastatin on endothelial cell function. Methods: 15 healthy persons and 30 patients with coronary heart disease were selected from June 2013 to June 2014 in our hospital. They were divided into healthy control group, coronary heart disease intervention group and coronary heart disease group. The results of blood enzyme linked immunosorbent assay (ELISAs) of asymmetric dimethyl arginine (ADMAN) and nitric oxide synthase (NOS) were compared in each group. Human umbilical vein endothelial cells (HUVECs) were cultured for 4 h and 8 h. According to the concentration of resuvastatin, they were divided into 5 groups: 0 渭 mol/L control group and 0.1 渭 mol/L 10 渭 mol/L group. To detect the expression of Risuvastatin in endothelial cells, the level of ADMA in the drug intervention group was significantly lower than that in the coronary heart disease group. But the levels of no and NOS were significantly higher than those of the healthy control group, but still lower than that of the healthy control group. The difference was statistically significant (P 0.05). The OD value of enzymatic marker was positively correlated with ADMA-no and NOS. The results of reverse transcription-polymerase chain reaction (RT-PCRR) showed that the expression of DDAH m RNA was increased in the group of 0.1 渭 mol/L and 100 渭 mol/L resuvastatin. The expression of DDAH m RNA in the 10 渭 mol/L and 100 渭 mol/L resuvastatin groups was significantly higher than that in the control group (P 0.05). The results of RT-PCR culture for 8 h showed that the DDAH m RNA expression in the 10 渭 mol/L and 100 渭 mol/L resuvastatin groups was increased. The expression of DDAH m RNA in the 10 渭 mol/L resuvastatin group was significantly higher than that in the control group (P 0.05). Conclusion: rosuvastatin has a better protective effect on endothelial cells and deserves clinical attention.
