陽離子交換-高效液相色譜-電化學(xué)法測定人血漿中兒茶酚胺的濃度

【摘要】:目的:采用弱陽離子固相萃取小柱(WCX-SPE)優(yōu)化人血漿兒茶酚胺檢測實驗條件,以減少血漿用量,提高實驗穩(wěn)定性,建立操作簡單、可靠的人血漿兒茶酚胺濃度檢測方法。方法:采用waters WCX固相萃取小柱提取血漿中兒茶酚胺,采用高效液相色譜-電化學(xué)檢測法測定兒茶酚胺,并對其線性范圍、方法回收率、提取回收率、日間日內(nèi)精密度及穩(wěn)定性進(jìn)行考察。結(jié)果:去甲腎上腺素(NE)在50~2 000 pg·ml-1(n=6,r=0.998 5),腎上腺素(E)在50~2 000 pg·ml-1(n=6,r=0.999 3)濃度范圍內(nèi)線性關(guān)系良好,最低檢測限分別為25,50 pg·ml-1(S/N≥3),NE、E樣品的平均提取回收率分別為89.6%、91.5%,平均方法回收率分別為100.6%,99.4%,平均日間RSD分別為5.1%,4.3%,平均日內(nèi)RSD分別為3.9%,3.6%。結(jié)論:采用WCX固相萃取小柱提取血漿中兒茶酚胺靈敏度高,操作簡便,結(jié)果準(zhǔn)確可靠,適合于臨床診療需要。
[Abstract]:Objective: to optimize the detection conditions of human plasma catecholamine by using weak cationic solid phase extraction column (WCX-SPE), in order to reduce the amount of plasma, improve the stability of the experiment, and establish a simple operation for the determination of catecholamine in human plasma. A reliable method for the determination of catecholamine in human plasma. Methods: the catecholamine in plasma was extracted by waters WCX solid-phase extraction column, and determined by high performance liquid chromatography-electrochemical detection. The recovery rate, intra-day precision and stability were investigated.
