南海紅樹(shù)林底泥放線(xiàn)菌Streptomyces sp.H41-26的抗菌化學(xué)成分研究

【摘要】:目的對(duì)分離自我國(guó)南海紅樹(shù)林底泥的鏈霉菌Streptomyces sp.H41-26菌株的發(fā)酵物抗菌活性成分進(jìn)行分離和鑒定。方法對(duì)鏈霉菌Streptomyces sp.H41-26菌株的發(fā)酵菌絲體采用95%乙醇提取,醇提取物經(jīng)乙酸乙酯萃取獲得抗菌活性部位,通過(guò)ODS開(kāi)放柱層析分離和反相高效液相制備純化。結(jié)果分離得到4個(gè)化合物,經(jīng)NMR和MS等波譜學(xué)方法分別鑒定為:放線(xiàn)菌素D(1)、放線(xiàn)菌素X_2(2)、放線(xiàn)菌素X_β(3)和放線(xiàn)菌素X_a(4)。結(jié)論化合物1~4在0.1mg/mL時(shí),對(duì)大腸埃希菌(ATCC8739)、金黃色葡萄球菌(ATCC6538P)、傷寒沙門(mén)菌(ATCC6539)、乙型副傷寒沙門(mén)菌(50094)和乙型溶血性鏈球菌(ATCC32204)具有明顯的抗菌活性,其中化合物2在0.01mg/mL時(shí),對(duì)除大腸埃希菌外的其他4種細(xì)菌抑制效果仍很明顯。
[Abstract]:Objective to isolate and identify the antimicrobial active components of Streptomyces Streptomyces sp.H41-26 from mangrove sediments in the South China Sea. Methods the fermentation mycelia of Streptomyces Streptomyces sp.H41-26 was extracted by 95% ethanol. Ethanol extract was extracted by ethyl acetate to obtain antibacterial active parts. The compounds were separated by ODS column chromatography and purified by reversed phase high performance liquid chromatography. The results of NMR and MS spectra were as follows: actinomycin D _ 1, actinomycin X _ 2, actinomycin X _ 尾 ~ (3) and actinomycin X _ 尾 ~ (3)), and actinomycin X _ 尾 ~ (3)) and actinomycin Xa4 ~ (4). Conclusion compound 1o _ 4 is at 0.1 mg / mL. It has obvious antimicrobial activity against Escherichia coli ATCC8739, Staphylococcus aureus ATCC6538PU, Salmonella typhimurium ATCC65399.Japanese paratyphoid Salmonella (50094) and B hemolytic streptococcus (ATCC32204), in which compound 2 is at 0.01 mg / mL. The inhibitory effect on the other four bacteria except Escherichia coli was still obvious.
