硒礦區(qū)土壤中真菌的分離培養(yǎng)及抗腫瘤活性篩選

【摘要】:利用單菌落挑選與劃線培養(yǎng)技術,從硒礦區(qū)土壤樣品中分離純化真菌。通過搖床發(fā)酵培養(yǎng),提取制備發(fā)酵樣品。采用MTT法,用MDA-MB-231細胞、Hela細胞、SMMC-7721細胞測試發(fā)酵樣品的抗腫瘤活性。從采自湖北恩施全球唯一獨立硒礦床區(qū)的6個不同土樣中分離真菌73株。其中,對人宮頸癌Hela細胞的IC_(50)小于90μg/m L的菌株32株,對人乳腺癌MDA-MB-231細胞的IC_(50)小于90μg/m L的菌株13株,對人肝癌SMMC-7721細胞的IC_(50)小于90μg/m L的菌株9株。這些活性菌株為篩選抗腫瘤天然產物提供了良好的藥源菌株。
[Abstract]:The fungi were isolated and purified from the soil samples of selenium mining area by single colony selection and underlined culture. The fermentation samples were extracted and prepared by shaking table fermentation. MTT method was used to isolate and purify the fungi from the soil samples. The antitumor activity of fermentative samples was tested by MDA-MB-231 cell line Hela cell line SMMC-7721. 73 strains of fungi were isolated from 6 different soil samples collected from the only independent selenium deposit area in Enshi, Hubei Province. For human cervical cancer Hela cells, 32 strains were less than 90 渭 g / mL, and 13 strains for human breast cancer MDA-MB-231 cells were less than 90 渭 g / mL. Nine strains of SMMC-7721 cells with less than 90 渭 g / mL were isolated from human hepatoma SMMC-7721 cells. These active strains provided a good drug source for screening natural anti-tumor products.
