牛溶菌肽線性衍生物Bac2a的生物特性及其穩(wěn)定性研究

【摘要】:本研究以牛中性粒細(xì)胞牛溶菌肽的線性衍生物Bac2a為研究對(duì)象,以蜂毒素為對(duì)照肽,探討B(tài)ac2a的生物特性及其穩(wěn)定性。采用圓二色譜法、微量稀釋法、噻唑藍(lán)(MTT)法等方法測(cè)定了Bac2a的二級(jí)結(jié)構(gòu)、抑菌活性、溶血活性、細(xì)胞毒性及其穩(wěn)定性。研究結(jié)果表明:Bac2a在水溶液中呈無(wú)規(guī)則卷曲狀,在疏水環(huán)境中則呈α-螺旋結(jié)構(gòu);Bac2a的抑菌作用較強(qiáng),最小抑菌濃度(MIC)為2~32μmol/L;當(dāng)肽濃度為64μmol/L時(shí),其溶血率較低,僅為14.81%;細(xì)胞毒性小,治療指數(shù)為3.26,細(xì)胞選擇性較強(qiáng);100℃處理1 h后,Bac2a依然保持較強(qiáng)的抑菌活性,熱穩(wěn)定性好。因此,Bac2a在食品、醫(yī)藥和其它領(lǐng)域都具有良好的應(yīng)用前景,有望成為抗生素替代品。
[Abstract]:In this study, the biological characteristics and stability of Bac2a, a linear derivative of bovine neutrophil bovine bacteriolytic peptide, and melittin as control peptide, were studied by circular dichroism chromatography and microdilution method. The secondary structure, bacteriostatic activity, hemolytic activity, cytotoxicity and stability of Bac2a were determined by thiazolyl blue method. In the hydrophobic environment, the bacteriostatic effect of Bac2a was stronger, the minimal inhibitory concentration was 2N 32 渭 mol / L, the hemolysis rate was lower when the concentration of peptide was 64 渭 mol/L, and the cytotoxicity was small.
