枸櫞酸坦度螺酮對(duì)自發(fā)性高血壓大鼠血壓的影響及機(jī)制研究

【摘要】:目的:研究枸櫞酸坦度螺酮(TC)對(duì)自發(fā)性高血壓大鼠(SHR)血壓的影響及機(jī)制。方法:將SHR隨機(jī)分為模型組、陽性對(duì)照組(苯磺酸左旋氨氯地平片,2.5 mg/kg)和TC高、中、低劑量組(TC膠囊,40、20、10 mg/kg),每組8只,另設(shè)8只正常大鼠為正常對(duì)照組,每天ig藥物1次,連續(xù)28 d。測(cè)量各組大鼠首次給藥前及給藥后0.5、1、1.5、2、3、4 h的收縮壓(SBP),另每給藥7 d測(cè)量1次1 h的SBP;末次給藥后檢測(cè)血清中一氧化氮(NO)含量,血漿中內(nèi)皮素、腎素、血管緊張素Ⅱ(AngⅡ)和去甲腎上腺素(NE)含量。結(jié)果:單次給藥時(shí),與模型組比較,陽性對(duì)照組和TC各劑量組SHR首次給藥后0.5、1 h時(shí)的SBP均明顯降低(P0.05或P0.01),之后僅陽性對(duì)照組和TC高劑量組的SBP明顯降低(P0.05或P0.01);多次給藥時(shí),與模型組比較,陽性對(duì)照組和TC高、中劑量組SHR給藥第7、14、28天的SBP均明顯降低(P0.05或P0.01)。與模型組比較,陽性對(duì)照組和TC高劑量組SHR血清中NO含量明顯升高,血漿中內(nèi)皮素、腎素、AngⅡ和NE含量均明顯降低(P0.05或P0.01);與陽性對(duì)照組比較,TC各劑量組SHR血清中NO含量明顯降低,TC中劑量組SHR血漿中內(nèi)皮素含量明顯升高(P0.05)。結(jié)論:TC可明顯降低SHR血壓,其機(jī)制可能與調(diào)節(jié)NO和內(nèi)皮素釋放平衡,降低腎素、AngⅡ及NE含量有關(guān)。
[Abstract]:Objective: to study the effect and mechanism of tandoxone citrate on blood pressure in spontaneously hypertensive rats. Methods: SHR was randomly divided into model group, positive control group (L-amlodipine benzenesulfonic acid 2.5 mg / kg) and high TC. In the low dose group, there were 8 rats in each group, 8 rats in each group, and 8 normal rats as control group.
