索拉膠對(duì)皮膚的保護(hù)作用研究

【摘要】:作為身體最大的器官,皮膚是身體抵御外界侵害的第一道防線,具有許多重要的作用。受到自身和外界環(huán)境的影響,皮膚逐漸出現(xiàn)干燥、發(fā)黃、松弛、暗淡、皺紋等現(xiàn)象。β-(1,3)-葡聚糖作為一種生物大分子,因具有高粘度、保濕、防皺、抗衰老和促進(jìn)傷口愈合等生物學(xué)活性,而被應(yīng)用于化妝品中,但由于β-(1,3)-葡聚糖的提取和純化復(fù)雜,使得其價(jià)格昂貴,難以被廣泛應(yīng)用。因此,研究水溶性的胞外p-(1,3)-葡聚糖—索拉膠在保濕、防皺、抗紫外和傷口愈合方面的性能,對(duì)于更好的利用和發(fā)揮索拉膠在化妝品和傷口愈合中的作用有著重要的意義。 在相對(duì)濕度為43%和81%的環(huán)境中,索拉膠的吸濕率分別為13.23%和20.07%;在相對(duì)濕度為43%的環(huán)境中,索拉膠的保濕率為23.93%,轉(zhuǎn)移至硅膠密閉容器里12小時(shí),索拉膠的保濕率為15.10%。人體皮膚保濕實(shí)驗(yàn)中,與處理前皮膚的含水率相比,索拉膠處理后的皮膚含水量增加百分比為82.05%,8小時(shí)后為43.58%。在成纖維細(xì)胞中,索拉膠加強(qiáng)了Collα1和Col3α1基因的表達(dá)。 在紫外線UVA照射前、后用索拉膠處理成纖維細(xì)胞3小時(shí),可以顯著提高細(xì)胞的存活率,與照射組的細(xì)胞存活率67.93%相比,照射前、后處理索拉膠的細(xì)胞存活率為100.94%、97.88%。小鼠實(shí)驗(yàn)中,索拉膠顯著降低了紫外照射引起的細(xì)胞因子IL-1β、TNF-α和IL-6的表達(dá),提高了Collα1和Co13α1基因的表達(dá),組織切片顯示,索拉膠可以削弱紫外線對(duì)皮膚中膠原纖維的破壞作用,維持皮膚的正常形態(tài)。在體外,索拉膠可以有效清除超氧陰離子自由基、羥基自由基和DPPH自由基,其清除率分別為17.53%、60.33%和43.82%。 在小鼠全皮層傷口模型中,索拉膠加速了傷口的愈合,第12天其傷口愈合率為5.38%,而對(duì)照組的為24.76%;提高了細(xì)胞因子IL-1p和TNF-α基因在傷口愈合前期的表達(dá),縮短了其高表達(dá)的時(shí)間,減輕了IL-1β和TNF-α的持續(xù)高表達(dá)對(duì)傷口愈合的不利作用。索拉膠可以促進(jìn)成纖維細(xì)胞的增殖,與對(duì)照組相比,索拉膠的濃度為500μg/ml時(shí),其細(xì)胞增殖率為124.95%。 所有研究表明,索拉膠具有良好的吸濕和保濕性能,可以提高皮膚含水率,并具有潛在的防皺作用,減輕紫外線對(duì)皮膚的傷害作用,有效清除自由基,促進(jìn)傷口愈合,可應(yīng)用于化妝品和傷口愈合類藥物等領(lǐng)域。
[Abstract]:As the largest organ of the body, skin is the body's first line of defense against external aggression and has many important functions. Under the influence of itself and the external environment, the skin gradually appears dry, yellow, relaxed and dark. Wrinkle and other phenomena. As a kind of biological macromolecule, 尾 -Che _ 1O _ 3C _ (- glucan) has been used in cosmetics because of its high viscosity, moisturizing, anti-wrinkle, anti-aging and promoting wound healing, but its extraction and purification are complicated. Therefore, to study the properties of water-soluble extracellular p-P- (1 +) -dextran-Sorghan in moisturizing, anti-wrinkle, anti-ultraviolet, and wound healing properties, which make it expensive and difficult to be widely used. It is of great significance to better use and play the role of Solan in cosmetic and wound healing. In the environment with relative humidity of 43% and 81%, the hygroscopic rates of sorghum were 13.23% and 20.07, respectively, and in the environment of 43% relative humidity, the moisture retention rate of sorghum was 23.93 and transferred to the silica gel closed container for 12 hours. In the skin moisturizing experiment, compared with the moisture content of the skin before the treatment, the moisture content of the skin treated with the solan gel increased by 82.05% and 43.58% after 8 hours. In fibroblasts, Solactin enhanced the expression of Coll 偽 1 and Col3 偽 1 genes. The survival rate of fibroblasts treated with Sola gel for 3 hours after ultraviolet UVA irradiation was significantly increased compared with that of irradiation group (67.93%). The cell survival rate of post-treated solan was 100.94% and 97.88%, compared with that of irradiation group (67.93%). Sola significantly decreased the expression of IL-1 尾 -TNF- 偽 and IL-6, and increased the expression of Coll 偽 1 and Co13 偽 1 genes. In vitro, Sola gum could effectively scavenge superoxide anion radical, hydroxyl radical and DPPH radical, and the scavenging rates were 17.53% and 43.82%, respectively. The wound healing rate was 5.38 on the 12th day and 24.76 in the control group, which increased the expression of cytokines IL-1p and TNF- 偽 in the early stage of wound healing and shortened the time of high expression. The continuous high expression of IL-1 尾 and TNF- 偽 could promote the proliferation of fibroblasts. Compared with the control group, the proliferation rate was 124.95% at the concentration of 500 渭 g / ml of IL-1 尾 and TNF- 偽. All the studies have shown that Sola gum has good moisture absorption and moisture retention, can increase moisture content of skin, and has potential anti-wrinkle effect, reduces the harm of ultraviolet rays to the skin, effectively clears free radicals, and promotes wound healing. Can be used in cosmetics and wound healing drugs and other fields.
