【摘要】：目的建立高效液相色譜-質譜聯(lián)用法(HPLC-MS/MS)定量測定人血漿中比卡魯胺的濃度。方法用蛋白沉淀處理血漿,以比卡魯胺-d4為內標,色譜柱:Agilent ZORBAX SB-C18柱(2.1 mm×50.0 mm,3.5μm),柱溫:20℃,流動相:甲醇-0.1%甲酸+2 mmol·L-1甲酸銨水溶液,等度洗脫,流速:0.35m L·min~(-1)。用負離子掃描模式,多反應監(jiān)測方式測定樣品中藥物的質量濃度。考察該方法的專屬性、標準曲線與定量下限、精密度與回收率、基質效應和穩(wěn)定性。結果血漿樣品中比卡魯胺回歸方程為y=6.82×10~(-3)x+1.33×10~(-2)(r=0.999 1)。比卡魯胺在10~1500 ng·mL~(-1)內線性關系良好,定量下限為10ng·mL~(-1)。日內、日間精密度的RSD均15%,平均回收率95%,穩(wěn)定性較好。結論本方法簡便快速、特異性強、靈敏準確,適用于人體血漿中比卡魯胺濃度的測定。
