【摘要】：目的:建立癲癇患者血漿中丙戊酸鈉(VPA)代謝產(chǎn)物2-丙基-2-戊烯酸(2-ene-VPA)的HPLC測(cè)定方法。方法:血漿用乙酸乙酯提取,以2,4’-二溴苯乙酮作為內(nèi)標(biāo),氮?dú)鈸]干樣品,色譜柱:ZORBAX SB-C18柱(4.6 mm×150 mm,5μm);流動(dòng)相:甲醇-水(55∶45);檢測(cè)波長(zhǎng):217 nm;流速:1.0ml·min-1;柱溫:35℃;進(jìn)樣溫度:20℃。結(jié)果:內(nèi)標(biāo)2,4’-二溴苯乙酮和2-ene-VPA的保留時(shí)間分別為11.2 min和5.1 min,2-ene-VPA的血漿濃度在0.5~16.0μg·ml-1范圍內(nèi),2-ene-VPA與內(nèi)標(biāo)2,4’-二溴苯乙酮的峰面積比與濃度呈良好的線性關(guān)系(R0.999),平均回收率為(96.85±0.33)%,日內(nèi)RSD5%,日間RSD5%。結(jié)論:本法經(jīng)濟(jì)、快速、簡(jiǎn)便,適用于臨床上2-ene-VPA的測(cè)定及研究。
[Abstract]:Objective: to establish a HPLC method for the determination of 2-propyl-2-pentene-VPA, a metabolite of sodium valproate in the plasma of epileptic patients.
