【摘要】：目的篩選西羅莫司自微乳給藥系統(tǒng)處方,并制備微丸。方法通過溶解度試驗確定助乳化劑、油相和乳化劑的選擇范圍;采用三元相圖法、星點設(shè)計和效應(yīng)面法對該體系優(yōu)化制備工藝及處方。采用擠出-滾圓法制備不同處方的西羅莫司自微乳化微丸。結(jié)果西羅莫司自微乳微丸的最終處方為:西羅莫司0.4%、油酸聚乙二醇甘油酯9.3%、聚氧乙烯-35-蓖麻油15.9%、二乙二醇單乙基醚8.0%、微晶纖維素49.8%、乳糖13.3%、羧甲基淀粉鈉3.3%。溶出度試驗顯示,西羅莫司固體自微乳微丸在水中的溶出度遠大于市售西羅莫司片,在0.4%SDS溶液中,兩制劑的溶出度相當(dāng)。結(jié)論自微乳化給藥系統(tǒng)可用于提高西羅莫司的體外溶出度。
[Abstract]:Objective to screen the prescription of sirolimus self-microemulsion delivery system and prepare pellets. Methods the selection range of co-emulsifier, oil phase and emulsifier was determined by solubility test, and the ternary phase diagram method was used. Star design and effect surface method were used to optimize the preparation process and formulation of the system. Different formulations of sirolimus self-emulsified pellets were prepared by extrusion-roller method. Results the final formulation of sirolimus self-emulsion pellets was as follows: sirolimo. The amount of polyethylene glycol oleate was 9.3%, polyoxyethylene 35 castor oil 15.9%, diethylene glycol monoethyl ether 8.0, microcrystalline cellulose 49.8%, lactose 13.3%, carboxymethyl starch sodium 3.3%. Dissolution test showed that, The dissolution of sirolimus solid self-emulsion pellets in water was much higher than that of sirolimus tablets sold on the market. In 0.4SDS solution, the dissolution rates of the two preparations were similar. Conclusion the self-microemulsified drug delivery system can be used to improve the dissolution of sirolimus in vitro.
