葉酸對PCPA抑郁小鼠外側(cè)韁核β-CaMKⅡ水平的影響

【摘要】:為探究葉酸對PCPA抑郁癥模型小鼠的干預(yù)作用及其對外側(cè)韁核β-Ca MK II水平的影響,將昆明種小鼠40只隨機分為三組:對照組、模型組、葉酸組,通過腹腔注射PCPA(100 mg/kg)每日1次,連續(xù)4 d構(gòu)建抑郁癥模型,曠場與懸尾實驗觀察小鼠行為變化,HPLC檢測小鼠下丘腦5-HT含量,免疫組化測定小鼠外側(cè)韁核β-Ca MK II表達水平。結(jié)果顯示,模型組與對照組相比自主活動能力下降,懸尾不動時間延長(P0.05),而葉酸組與對照組相比上述實驗結(jié)果無統(tǒng)計學差異(P0.05)。模型組與對照組相比下丘腦5-HT含量降低,外側(cè)韁核β-Ca MK II表達水平下降(P0.05)。結(jié)果表明葉酸能顯著提高下丘腦5-HT含量與外側(cè)韁核β-Ca MK II水平,其抗抑郁作用機制可能與之有關(guān)。
[Abstract]:In order to investigate the effect of folic acid on PCPA depression model mice and the effect of folic acid on the level of 尾 -Ca MK II in lateral habenula, 40 Kunming mice were randomly divided into three groups: control group and model group. In folic acid group, the depression model was established by intraperitoneal injection of PCPA(100 mg / kg once a day for 4 days. The behavior of mice was observed by open-field and tail suspension experiments. The content of 5-HT in hypothalamus and the expression of 尾 -CaMKII in lateral habenular nucleus of mice were detected by HPLC and immunohistochemistry. The results showed that the ability of autonomous activity in model group was lower than that in control group. The immobility time of tail was prolonged (P0.05A), but there was no statistical difference between the folic acid group and the control group (P 0.05). The content of 5-HT in hypothalamus of model group was lower than that of control group. The expression level of 尾 -CaMK II in the lateral habenular nucleus decreased (P 0.05). The results showed that folic acid could significantly increase 5-HT content in the hypothalamus and 尾 -Ca MK II level in the lateral habenular nucleus. Its antidepressant mechanism may be related to it.
