【摘要】:目的:建立人單核細(xì)胞THP-1源性泡沫細(xì)胞內(nèi)雷帕霉素(RAPA)含量的測(cè)定方法,研究RAPA靶向制劑(RAPA-NP-Apt)對(duì)泡沫細(xì)胞的靶向作用。方法:以氧化型低密度脂蛋白誘導(dǎo)THP-1細(xì)胞建立泡沫細(xì)胞模型,取200 ng/m L的RAPA和200、400、800ng/m L的RAPA-NP-Apt與泡沫細(xì)胞共同孵育60 min。采用高效液相色譜法(HPLC)測(cè)定細(xì)胞內(nèi)RAPA含量,色譜柱為Diamonsil C18,流動(dòng)相為乙腈-水(90∶10,V/V),流速為1.0 m L/min,柱溫為40℃,檢測(cè)波長(zhǎng)為278 nm,進(jìn)樣量為20μL。結(jié)果:RAPA檢測(cè)質(zhì)量濃度線性范圍為50~6 400 ng/m L(r=0.999 96),平均回收率為98.72%(RSD=0.62%,n=3),日內(nèi)、日間RSD均不大于6.15%(n=6),穩(wěn)定性試驗(yàn)的RSD2%(n=6),重復(fù)性試驗(yàn)的RSD=1.64%(n=6)。RAPA和低、中、高質(zhì)量濃度的RAPA-NP-Apt與泡沫細(xì)胞共孵育后RAPA的含量分別為12、43、98、140 ng/10~6個(gè)細(xì)胞。結(jié)論:本方法操作簡(jiǎn)單,穩(wěn)定性和重復(fù)性好,可用于泡沫細(xì)胞內(nèi)RAPA的含量測(cè)定。RAPA-NP-Apt可提高RAPA對(duì)泡沫細(xì)胞的靶向作用。
