【摘要】：考察了膦甲酸鈉對(duì)硫酸頭孢噻利在大鼠體內(nèi)藥動(dòng)學(xué)的影響。健康Wistar大鼠隨機(jī)分為試驗(yàn)組與對(duì)照組,每組20只。試驗(yàn)組大鼠尾靜脈注射膦甲酸鈉360 mg/kg,隨后即刻注射硫酸頭孢噻利200 mg/kg;對(duì)照組僅注射硫酸頭孢噻利200 mg/kg。采用HPLC法測(cè)定血漿中硫酸頭孢噻利濃度,并擬合藥動(dòng)學(xué)參數(shù),試驗(yàn)組和對(duì)照組結(jié)果如下:t1/2(1.76±0.57)和(1.22±0.51)h,cmax(1 314.37±196.70)和(1 470.18±321.38)μg/ml,AUC0→24 h(863.19±189.46)和(799.57±203.39)μg·h·ml-1,AUC0→∞(898.81±214.25)和(800.02±207.04)μg·h·ml-1,V(0.56±0.18)和(0.45±0.17)L/kg,CL(0.23±0.05)和(0.27±0.07)L·h-1·kg-1。合用膦甲酸鈉后,硫酸頭孢噻利在大鼠體內(nèi)的t1/2和V顯著升高,表明膦甲酸鈉與硫酸頭孢噻利可能競(jìng)爭(zhēng)同一排泄途徑,減緩硫酸頭孢噻利的排泄。
[Abstract]:The effect of phosphonium formate on the pharmacokinetics of ceftithione sulfate in rats was investigated. Healthy Wistar rats were randomly divided into two groups: the experimental group and the control group. 20 rats in each group were injected with sodium phosphonate 360 mg / kg via caudal vein and then 200 mg / kg ceftiazil sulfate was injected immediately afterwards. The control group was only injected with ceftiazol sulfate 200mg / kg. The plasma ceftithiazole sulfate concentration was determined by HPLC method and the pharmacokinetic parameters were fitted.
