經(jīng)導(dǎo)管射頻消融去腎交感神經(jīng)術(shù)對高血壓犬心肌纖維化及miR-133a/CTGF的影響

目的:觀察經(jīng)導(dǎo)管射頻消融去腎交感神經(jīng)術(shù)(RDN)對高血壓犬心肌纖維化程度、心肌miR-133a、結(jié)締組織生長因子(CTGF)表達的改變,探討RDN對心肌纖維化的影響及可能的機制。方法:1、用高脂飲食建立高血壓犬模型,將28只比格犬(10-12月齡)隨機分為對照組(普通飲食,n=6)和模型組(高脂飲食,n=22)。2、建模12周后將建模成功的高血壓犬(n=20)隨機分為手術(shù)組(n=10)及假手術(shù)組(n=10),手術(shù)組行RDN術(shù),假手術(shù)組僅行腎動脈造影。3、監(jiān)測實驗犬體重、血壓,術(shù)后6月無痛處死實驗犬,取心臟組織,行Masson染色觀察心肌纖維化程度,計算膠原容積分數(shù)(CVF)、周圍血管膠原面積比率(PVCA), Real-time PCR檢測組織miR-133a表達水平;Real-time PCR、Western-blot檢測組織CTGF表達。結(jié)果:1、建模期假手術(shù)組、手術(shù)組實驗犬體重、收縮壓(SBP)、舒張壓(DBP)、平均動脈壓(MAP)均明顯升高,與基線、對照組相比均有統(tǒng)計學(xué)差異(P<0.05),假手術(shù)組、手術(shù)組組間對比無統(tǒng)計學(xué)差異(P>0.05);2、經(jīng)導(dǎo)管射頻消融去...
