【摘要】：目的建立一種損傷更小、存活率更高、適用范圍更廣的改良的脊髓半橫斷損傷大鼠模型。方法將20只雌性SpragueDawley大鼠隨機分為常規(guī)脊髓半橫斷模型組和改良脊髓半橫斷模型組,分別采用常規(guī)和改良的脊髓半橫斷法制備大鼠脊髓損傷模型,分別于術(shù)后1 d、3 d、1周和2周通過Basso-Beattie-Bresnahan(BBB)運動功能學(xué)評分評價大鼠后肢運動功能的恢復(fù)情況,并結(jié)合手術(shù)時間、術(shù)中出血量、術(shù)中脊髓后正中線定位以及二次手術(shù)顯露脊髓的成功率等主要參數(shù),比較2組間的差異。結(jié)果改良脊髓半橫斷模型組較常規(guī)脊髓半橫斷模型組手術(shù)時間更短,術(shù)中出血量明顯減少,BBB運動功能學(xué)評分更優(yōu),術(shù)中脊髓后正中線定位更準確,二次手術(shù)顯露脊髓的成功率更高。在改良的脊髓半橫斷模型基礎(chǔ)上,成功建立了脊髓瘢痕組織移植模型,成功率可達70%以上。結(jié)論改良的脊髓半橫斷模型在手術(shù)時間、術(shù)中出血量、運動功能學(xué)評分、術(shù)中脊髓后正中線定位及二次手術(shù)顯露脊髓方面均表現(xiàn)出明顯優(yōu)勢,更適用于脊髓損傷研究,尤其是非急性期脊髓損傷研究。
[Abstract]:Objective to establish a modified rat model of spinal cord hemitransection injury with less injury, higher survival rate and wider application range. Methods Twenty female SpragueDawley rats were randomly divided into two groups: the normal spinal cord hemitransection model group and the modified spinal cord hemitransection model group. Spinal cord injury models were established by routine and modified hemitransection of spinal cord in rats. The recovery of motor function of hind limbs was evaluated by Basso-Beattie-Bresnahanna BBB score at 1 and 2 days after operation, and combined with operation time. The main parameters, such as intraoperative bleeding, localization of spinal cord posterior median line, and the success rate of second operation, were compared between the two groups. Results the operation time of the modified spinal cord hemitransection model group was shorter than that of the conventional spinal cord hemitransection model group. The intraoperative bleeding volume was significantly reduced and the motor function score of BBB was significantly reduced. The localization of posterior median line of spinal cord was more accurate, and the success rate of exposing spinal cord by secondary operation was higher. Based on the improved model of hemitransection of spinal cord, The successful rate of spinal cord scar transplantation was over 70%. Conclusion the modified spinal cord hemitransection model has the advantages of operation time, intraoperative blood loss, motor function score and so on. The localization of posterior median line of spinal cord and the exposure of spinal cord by secondary operation showed obvious advantages, and were more suitable for the study of spinal cord injury, especially for non-acute spinal cord injury.
