血管生物反應(yīng)器的搭建及可降解鎂基支架細(xì)胞應(yīng)答的研究

生物可降解鎂基金屬材料在心血管疾病介入治療中被譽(yù)為最有潛力的可降解血管支架替代材料。在鎂基支架的設(shè)計(jì)與研發(fā)過(guò)程中發(fā)展了許多體外評(píng)價(jià)鎂基材料的生物相容性及其降解行為的方法。但是這些評(píng)價(jià)方法存在不足,主要表現(xiàn)為目前的方法無(wú)法全面地揭示細(xì)胞對(duì)材料降解的影響以及細(xì)胞自身對(duì)材料降解的響應(yīng);此外材料本身降解行為在體內(nèi)外存在很大差異,體外評(píng)價(jià)結(jié)果不能很好地為支架在體內(nèi)甚至臨床實(shí)驗(yàn)提供參考或者指導(dǎo)。本研究針對(duì)上述問(wèn)題成功構(gòu)建了具有從結(jié)構(gòu)和功能上高度模擬血管微環(huán)境的血管生物反應(yīng)器,對(duì)植入鎂基支架的人臍帶動(dòng)脈和兔頸動(dòng)脈血管在動(dòng)態(tài)的生物反應(yīng)器中和靜態(tài)條件下進(jìn)行培養(yǎng),結(jié)果表明:動(dòng)態(tài)的流體環(huán)境比傳統(tǒng)靜態(tài)條件更適于維持血管組織在體外的活性,并且支架降解產(chǎn)物可得到?jīng)_刷,降解引起的pH變化也可以得到實(shí)時(shí)緩沖。此外,通過(guò)設(shè)計(jì)兔頸動(dòng)脈血管支架植入實(shí)驗(yàn)與基于血管生物反應(yīng)器的支架-血管實(shí)驗(yàn)作對(duì)比,以評(píng)估該血管生物反應(yīng)器的實(shí)際效用。結(jié)果表明:支架引起的細(xì)胞組織應(yīng)答在體內(nèi)和體外生物反應(yīng)器中依然存在差異,在動(dòng)物體內(nèi)新生內(nèi)膜容易覆蓋支架筋周?chē)?而在體外血管生物反應(yīng)器中卻難以實(shí)現(xiàn)這一點(diǎn)。因此生物反應(yīng)器的培養(yǎng)條件還需進(jìn)一步改進(jìn),使其能更...
