自組裝肽納米纖維水凝膠支架對TGF的緩釋作用及其生物學特性的實驗研究

第一部分自組裝肽納米纖維水凝膠支架材料的制備及理化性質(zhì)分析目的設計并制備由Arg、Ala、Asp等氨基酸構(gòu)成的新型自組裝肽(self-assemblingpeptide,SAP):L-RADA16、D-RADA16 和 L-RADA16-RGD。采用圓二色譜儀、掃描電鏡、透射電鏡、流變儀對L-RADA16、D-RADA16和L-RADA16-RGD進行表征,了解其微觀結(jié)構(gòu),評估其是否可形成納米纖維支架結(jié)構(gòu);研究三者的機械性質(zhì),比較其物理學穩(wěn)定性。以期研發(fā)兼具良好穩(wěn)定性與生物學活性,同時可批量化生產(chǎn)的骨修復材料。方法三種SAP均采用商業(yè)合成,經(jīng)高效液相色譜儀(high performance liquid chromatography,HPLC)分析純度;采用圓二色譜(circular dichroism,CD)了解三種 SAP L-RADA16、D-RADA16 及 L-RADA16-RGD 的二級結(jié)構(gòu);采用掃描電鏡(scanning electron microscopy,SEM)和透射電鏡(transmission electron microscopy,TEM)觀察三種 SAP ...
