第一部分體外誘導(dǎo)骨髓間充質(zhì)干細(xì)胞（Bone marrow mesenchymalstem cells,BMSCs）定向分化為內(nèi)皮細(xì)胞、平滑肌細(xì)胞、上皮細(xì)胞和成纖維細(xì)胞目的:探索體外誘導(dǎo)BMSCs向內(nèi)皮細(xì)胞、平滑肌細(xì)胞、上皮細(xì)胞和成纖維細(xì)胞分化的方法,為3D生物打印法重建陰道提供種子細(xì)胞。方法:體外培養(yǎng)大鼠BMSCs1 CoCl₂模擬乏氧微環(huán)境:MTT法檢測CoCl₂對BMSCs生長的影響,Western blot法檢測Co Cl₂作用后HIF-1α的表達(dá)情況,鑒定CoCl₂模擬乏氧微環(huán)境的能力。2誘導(dǎo)BMSCs向內(nèi)皮細(xì)胞、平滑肌細(xì)胞、上皮細(xì)胞和成纖維細(xì)胞分化:內(nèi)皮細(xì)胞誘導(dǎo):CoCl₂ 100μM預(yù)處理BMSCs 24 h體外模擬乏氧微環(huán)境,更換為內(nèi)皮誘導(dǎo)培養(yǎng)基（3%FBS,50 ng/mL VEGF和10 ng/mL bFGF）誘導(dǎo)細(xì)胞7/14天。平滑肌細(xì)胞誘導(dǎo):平滑肌細(xì)胞誘導(dǎo)培養(yǎng)基（10%FBS,10 ng/mL PDGF-AB和5 ng/mL TGF-β1）誘導(dǎo)BMSCs ...
