結(jié)直腸癌患者血液表面增強(qiáng)拉曼光譜研究

【摘要】:表面增強(qiáng)拉曼光譜技術(shù)(SERS)是當(dāng)今國際生物醫(yī)學(xué)領(lǐng)域的熱點(diǎn)研究技術(shù),本論文根據(jù)結(jié)直腸癌臨床診斷需求,將具有高生物檢測(cè)靈敏度的SERS技術(shù)應(yīng)用于結(jié)直腸癌患者血液的分析,并結(jié)合醫(yī)學(xué)統(tǒng)計(jì)方法,試圖發(fā)展一種無損、快速、客觀、準(zhǔn)確的基于SERS技術(shù)的結(jié)直腸癌血液檢測(cè)方法。主要研究工作包括: 1.研究以銀納米顆粒為增強(qiáng)基底的結(jié)直腸癌病人血液的SERS光譜,通過對(duì)比38個(gè)結(jié)直腸癌患者與45個(gè)健康人的血液SERS光譜并結(jié)合多元統(tǒng)計(jì)方法,對(duì)二者的光譜進(jìn)行區(qū)分與判別,診斷靈敏度和特異性分別為81.6%和75.6%。 2.將蛋白膜電泳技術(shù)與基于銀納米顆粒的SERS技術(shù)相結(jié)合,試圖從血液蛋白SERS的角度探索結(jié)直腸癌檢測(cè)的可能性。結(jié)果顯示:利用血清白蛋白SERS光譜來區(qū)分結(jié)直腸癌患者與健康人的診斷方法的靈敏度和特異性分別為84.2%和80%;而血清球蛋白則為84.2%和75.6%。結(jié)果表明:電泳提純的血清蛋白的診斷效果優(yōu)于全血清SERS的診斷效果,進(jìn)一步提高了血液SERS的結(jié)直腸癌診斷效果。 3.提出了一種利用接收光偏振SERS技術(shù)檢測(cè)結(jié)直腸癌血液的新方法。判別結(jié)果顯示:當(dāng)接收光路上的偏振片豎直(0°)、水平(90°)放置時(shí),結(jié)直腸癌血清的檢測(cè)靈敏度分別為89.5%和100%;特異性分別為93.3%和84.4%。結(jié)果表明:基于接收光偏振SERS技術(shù)的結(jié)直腸癌血清診斷效果明顯優(yōu)于常規(guī)SERS的診斷效果。 4.根據(jù)金納米顆粒更具生物兼容性的特點(diǎn),研究以金納米顆粒為增強(qiáng)基底的結(jié)直腸癌病人血液的SERS光譜,通過對(duì)比結(jié)直腸癌患者與健康人的血液SERS光譜并結(jié)合多元統(tǒng)計(jì)方法,對(duì)二者的光譜進(jìn)行區(qū)分與判別,診斷靈敏度和特異性分別達(dá)到97.4%和100%,診斷效果要明顯優(yōu)于銀膠SERS。結(jié)果表明:利用基于金納米粒子的SERS技術(shù)并結(jié)合LDA-PCA的光譜判別診斷法對(duì)癌癥血清進(jìn)行分析,是一種新穎的、無損的、極具潛力的結(jié)直腸癌檢測(cè)及篩選手段。
[Abstract]:Surface enhanced Raman spectroscopy (SERS) is a hot research technique in the field of biomedicine. According to the requirement of clinical diagnosis of colorectal cancer, the SERS technique with high sensitivity of biological detection is applied to the analysis of the blood of colorectal cancer patients. Combined with the medical statistical method, this paper attempts to develop a nondestructive, rapid, objective and accurate blood detection method for colorectal cancer based on SERS technology. The main research work includes: 1. The SERS spectra of the blood of colorectal cancer patients with silver nanoparticles as the enhancement substrate were studied. By comparing the SERS spectra of 38 colorectal cancer patients with 45 healthy people and combining with the multivariate statistical method, the spectra of the two groups were distinguished and distinguished. The diagnostic sensitivity and specificity were 81.6% and 75.6%, respectively. In order to explore the possibility of detection of colorectal cancer from the point of view of blood protein SERS, the technique of protein membrane electrophoresis was combined with SERS technology based on silver nanoparticles. The results showed that the sensitivity and specificity of the method were 84.2% and 80%, respectively, and those of serum globulin were 84.2% and 75.6%, respectively. The results showed that the diagnostic effect of serum protein purified by electrophoresis was better than that of whole serum SERS, and further improved the diagnostic effect of blood SERS for colorectal cancer. A new method for detecting the blood of colorectal cancer by receiving light polarization SERS is proposed. The discriminant results showed that when the polarizer was placed vertically (0 擄) and horizontally (90 擄), the detection sensitivity was 89.5% and 100%, and the specificity was 93.3% and 84.4%, respectively. The results showed that the diagnostic effect of SERS based on receiving light polarization was significantly better than that of conventional SERS. 4. According to the more biocompatible characteristics of gold nanoparticles, the SERS spectra of the blood of colorectal cancer patients based on gold nanoparticles were studied. By comparing the SERS spectra of the patients with colorectal cancer and healthy people and combining the multivariate statistical method, The diagnostic sensitivity and specificity were 97.4% and 100% respectively, and the diagnostic effect was obviously better than that of silver gel SERS. The results showed that the SERS technique based on gold nanoparticles and the spectral discriminant diagnostic method based on LDA-PCA were a novel, non-destructive and potential method for detection and screening of colorectal cancer.
