軟骨細胞-海藻酸鈉水凝膠微球修復大鼠顱骨缺損

【摘要】:目的探討軟骨細胞-海藻酸鈉水凝膠修復骨缺損的可行性,比較其與單純海藻酸鈉水凝膠修復效果的差別。方法取SD大鼠乳鼠膝關節(jié)軟骨分離培養(yǎng)獲得第2代軟骨細胞,以1×106/m L接種于海藻酸鈉水溶液中培養(yǎng)并交聯(lián)形成復合水凝膠微球。取12只2月齡雄性SD大鼠,建立顱頂單側缺損模型,隨機分為實驗組與對照組,分別植入軟骨細胞-海藻酸鈉水凝膠和單純海藻酸鈉水凝膠微球。于術后6周取材行大體及組織學觀察以及Micro-CT掃描定量分析。結果大體觀察示術后6周實驗組缺損已基本修復完全,表面光滑無塌陷或碎裂,質地較硬接近新生骨組織;對照組缺損明顯,周圍為不規(guī)則骨和纖維結締樣組織。Micro-CT掃描示實驗組顱骨缺損得到良好的修復重建,定量分析示實驗組骨礦物質密度、骨組織體積分數(shù)及骨小梁數(shù)目均高于對照組,差異有統(tǒng)計學意義(P0.05)。組織學觀察示術后6周實驗組缺損區(qū)由新生骨組織修復,含少量軟骨及纖維結締組織,出現(xiàn)血管結構;對照組缺損明顯,邊緣主要由纖維結締組織和少量新生骨組織構成。結論海藻酸鈉復合軟骨細胞制備的水凝膠微球對于大鼠顱骨缺損具有一定的修復效果。
[Abstract]:Objective to investigate the feasibility of repairing bone defect by chondrocyte sodium alginate hydrogel and compare the effect of sodium alginate hydrogel alone. Methods the second generation of chondrocytes were obtained from the articular cartilage of neonatal SD rats. The compound hydrogel microspheres were cultured and crosslinked in sodium alginate solution with 1 脳 10 6 / mL of 1 脳 10 6 mL. Twelve 2-month-old male SD rats were used to establish a unilateral cranial defect model and were randomly divided into two groups: experimental group and control group. Chondrocyte sodium alginate hydrogel and sodium alginate hydrogel microspheres were implanted respectively. Gross and histological observation and quantitative analysis of Micro-CT scanning were performed 6 weeks after operation. The gross observation showed that the experimental group was deficient 6 weeks after operation. The damage has been basically repaired, There was no collapse or fragmentation on the smooth surface, and the texture was hard and close to the new bone tissue, while the defect in the control group was obvious, and the irregular bone and fibrous connective tissue. Micro-CT scanning showed that the skull defect in the experimental group was well repaired and reconstructed. Quantitative analysis showed that the bone mineral density, volume fraction of bone tissue and the number of trabeculae in the experimental group were higher than those in the control group, and the difference was statistically significant (P 0.05). Histological observation showed that the defect area of the experimental group was repaired by new bone tissue at 6 weeks after operation. There was a small amount of cartilage and fibrous connective tissue, and vascular structure appeared in the control group, but the defect was obvious in the control group. Conclusion the hydrogel microspheres prepared by sodium alginate combined with chondrocytes can repair the skull defect in rats.
