調(diào)節(jié)性T細(xì)胞對(duì)類風(fēng)濕關(guān)節(jié)炎治療作用的研究

研究背景:類風(fēng)濕關(guān)節(jié)炎(rheumatoid arthritis,RA)是一種系統(tǒng)性自身免疫性關(guān)節(jié)炎。表現(xiàn)為關(guān)節(jié)炎癥,滑膜增生,局部炎癥細(xì)胞浸潤(rùn),細(xì)胞因子異常分泌,關(guān)節(jié)軟骨和骨組織破壞等。RA患者關(guān)節(jié)滑膜成纖維細(xì)胞(rheumatoid arthritis synovial cells,RASF)表現(xiàn)出異常增殖和凋亡減低特性,對(duì)RA發(fā)病非常重要。近年,免疫學(xué)方面的研究發(fā)現(xiàn)調(diào)節(jié)性T細(xì)胞(regulatory Tcell,Treg)在RA發(fā)病過(guò)程中起重要作用。Treg細(xì)胞由CD4+T細(xì)胞亞群分化而來(lái),占CD4+T細(xì)胞的5%-10%。Treg細(xì)胞特異性高表達(dá)白細(xì)胞介素(interleukin,IL)-2受體a鏈CD25分子,并能分泌IL-10、IL-35和轉(zhuǎn)化生長(zhǎng)因子(transforming growth factor-β,TGF-β)等細(xì)胞因子,具有強(qiáng)大的免疫抑制功能。Treg細(xì)胞與RA發(fā)病密切相關(guān),RA患者外周血Treg細(xì)胞的數(shù)量和功能均存在缺陷,導(dǎo)致效應(yīng)性T細(xì)胞過(guò)度活化,促進(jìn)RA發(fā)病。輔助性T細(xì)胞(helper T cell,Th)17也來(lái)自CD4+T細(xì)胞,與Treg細(xì)胞功能相反,在...
