中央杏仁核GLP-1受體信號通路的攝食調(diào)控研究

目的:中央杏仁核是胰高血糖素樣肽1(GLP-1)攝食調(diào)控重要作用位點(diǎn)之一,中央杏仁核內(nèi)GLP-1受體信號通路在攝食和能量代謝調(diào)控中發(fā)揮著重要生理作用。本研究擬通過熒光免疫組化染色、核團(tuán)微量注射等方法,觀察激活或阻斷中央杏仁核內(nèi)GLP-1信號通路對大鼠攝食影響及潛在機(jī)制。方法:選用成年健康雄性Wistar大鼠,體重280-320 g。采用熒光免疫組織化學(xué)染色,觀察大鼠中央杏仁核中GLP-1受體的分布;采用腦核團(tuán)置管及中央杏仁核微量注射GLP-1受體激動劑Ex4或GLP-1受體拮抗劑Ex9,觀察GLP-1受體信號通路激活或阻斷對大鼠攝食量、高脂飲食、高嶺土攝入、以及蔗糖和玉米油攝入的影響。結(jié)果:(1)大鼠中央杏仁核和尾殼核分別注射6-羧基熒光素(FAM)標(biāo)記的Ex4(GLP-1受體激動劑),3小時后中央杏仁核內(nèi),而不是尾殼核,可見有呈黃色熒光的FAM,結(jié)合熒光免疫組化神經(jīng)元核(NeuN)染色,確認(rèn)FAM位于中央杏仁核神經(jīng)元內(nèi)。提示,GLP-1受體在中央杏仁核內(nèi)的表達(dá)具有位置特異性。(2)中央杏仁核微量注射GLP-1受體激動劑Ex4,大鼠0-20 h累計攝食量顯著降低(P<0.05);...
