IGFBP2通過ERK1/2信號通路調(diào)控RA-FLS細胞生物學行為機制研究

目的:探討胰島素樣生長因子結(jié)合蛋白2（Insulinlike growth factor binding protein2,IGFBP2）對類風濕關(guān)節(jié)炎成纖維樣滑膜細胞（RA-FLS）生物學行為的作用,以及IGFBP2對細胞外調(diào)節(jié)蛋白激酶（Extracellular regulated protein kinases1/2,ERK1/2）的作用。方法:首先,我們原代培養(yǎng)類風濕關(guān)節(jié)炎成纖維樣滑膜細胞（RA-FLS）和正常成纖維樣滑膜細胞（FLS）,并采用q RT-PCR、Western blot及免疫熒光技術(shù)檢測IGFBP2 m RNA和蛋白在FLS和RA-FLS中的表達水平。其次,RA-FLS細胞被轉(zhuǎn)染si RNA-IGFBP2和si RNA-NC,q RT-PCR檢測IGFBP2在RA-FLS細胞中的干擾效果;CCK-8實驗檢測IGFBP2敲低后對RA-FLS細胞增殖能力的影響;流式細胞儀檢測IGFBP2敲低后對RA-FLS細胞凋亡的影響;Transwell檢測IGFBP2敲低對RA-FLS細胞侵襲能力的影響;劃痕實驗檢測IGFBP2敲低對RA-FLS細胞遷移能力的影響。此外,RA-...
