mTOR通路對糖尿病雪旺氏細(xì)胞凋亡影響的研究

糖尿病周圍神經(jīng)。╠iabetic peripheral neuropathy,DPN)是糖尿病常見的慢性并發(fā)癥之一,損害周圍神經(jīng)的髓鞘和軸索,導(dǎo)致周圍神經(jīng)傳導(dǎo)速度減慢和動作電位波幅降低,造成患者出現(xiàn)一系列感覺、運動和自主神經(jīng)功能障礙,如:肢體麻木、疼痛、感覺異常甚至肌無力和肌肉萎縮,嚴(yán)重影響患者的生活質(zhì)量。Unger等人研究發(fā)現(xiàn)糖尿病大鼠有髓神經(jīng)纖維(腓腸神經(jīng))的橫切面積下降,主要源于髓鞘變薄,而非軸索直徑的改變。雪旺氏細(xì)胞參與了周圍神經(jīng)髓鞘的形成和再生,是維持周圍神經(jīng)生理功能的關(guān)鍵組分。雪旺氏細(xì)胞的損傷是引起糖尿病周圍神經(jīng)病發(fā)生的機制之一,在糖尿病動物模型中可檢測到雪旺氏細(xì)胞的凋亡,進(jìn)一步揭示葡萄糖和一些來源于葡萄糖的代謝產(chǎn)物,如丙酮醛(methylglyoxal,MG)和晚期糖基化終末產(chǎn)物(advanced glycation end products,AGEs)是導(dǎo)致糖尿病雪旺氏細(xì)胞凋亡的主要因素。細(xì)胞凋亡的發(fā)生可由多種途徑來介導(dǎo),包括線粒體介導(dǎo)的凋亡途徑、內(nèi)質(zhì)網(wǎng)應(yīng)激介導(dǎo)的凋亡途徑和死亡受體介導(dǎo)的凋亡途徑。線粒體介導(dǎo)的凋亡通過調(diào)控線粒體膜上的促凋亡分子(Bax)和抗凋亡分子(B...
