碘過量對(duì)胰島β細(xì)胞功能的影響及機(jī)制

【摘要】:目的探索碘過量暴露對(duì)小鼠胰島素瘤細(xì)胞系β-TC-6胰島素分泌功能影響及機(jī)制。方法β-TC-6細(xì)胞在對(duì)數(shù)生長(zhǎng)期暴露于0、0.01、0.1、1、10和100 mmol/L碘24 h,采用四氮唑鹽(MTT)比色法檢測(cè)細(xì)胞存活率。細(xì)胞經(jīng)0.01、1、和100 mmol/L碘干預(yù)24 h后,用5.6 mmol/L葡萄糖刺激細(xì)胞1 h,酶聯(lián)免疫分析(ELISA)法檢測(cè)上清胰島素含量。蛋白印跡法(Western blot)檢測(cè)GRP78、IRE1α、Bcl-2和Bax蛋白表達(dá)。結(jié)果與對(duì)照組比較,1~100 mmol/L碘處理組細(xì)胞存活率顯著降低(P0.05);1 mmol/L組細(xì)胞胰島素分泌量顯著增加(P0.05),100 mmol/L組細(xì)胞胰島素分泌量顯著下降(P0.05),1和10 mmol/L組GRP78、IRE1α和Bax蛋白表達(dá)量顯著增加(P0.05),而Bcl-2蛋白表達(dá)顯著降低(P0.05)。5.6 mmol/L葡萄糖+0 mmol/L碘組、5.6 mmol/L葡萄糖+0.01 mmol/L碘組和5.6 mmol/L葡萄糖+1 mmol/L碘組分別與相應(yīng)對(duì)照組比較,細(xì)胞胰島素分泌量組間差異顯著(P0.05)。結(jié)論過量碘可降低β-TC-6細(xì)胞胰島素分泌量,損害胰島素分泌功能,其機(jī)制很可能與內(nèi)質(zhì)網(wǎng)應(yīng)激和Bcl-2/Bax蛋白表達(dá)比例失衡密切相關(guān)。
