兒茶素和左甲狀腺素鈉對甲狀腺功能減退大鼠抗氧化應(yīng)激能力改善的影響

【摘要】:目的觀察左甲狀腺素鈉、兒茶素以及兩藥聯(lián)合治療對甲狀腺功能減退模型大鼠血清8-羥基脫氧鳥苷(8-OHd G)、對氧磷酶-1含量的影響以及海馬組織凋亡蛋白caspase-3含量的影響。方法選取50只成年SD雄性大鼠,隨機選取10只作為對照組,剩余40只用丙基硫氧密啶(PTU)誘導(dǎo)甲狀腺功能減退大鼠模型,分為PTU組、兒茶素組、左甲狀腺素鈉組(L-T4組)、左甲狀腺素鈉+兒茶素組(聯(lián)合用藥組)。采用酶聯(lián)免疫吸附試驗測定各組大鼠血清8-OHd G、對氧磷酶-1水平。采用Western blot測定各組大鼠海馬組織凋亡蛋白caspase-3的含量。結(jié)果 1與對照組比較,PTU組大鼠血清8-OHd G含量和海馬組織凋亡蛋白caspase-3含量顯著升高,差異均有統(tǒng)計學(xué)意義(P0.05);與PTU組比較,L-T4組、聯(lián)合用藥組大鼠血清8-OHd G含量和海馬組織凋亡蛋白caspase-3含量顯著降低,差異均有統(tǒng)計學(xué)意義(P0.05)。2與對照組比較,PTU組大鼠血清對氧磷酶-1含量顯著降低,差異有統(tǒng)計學(xué)意義(P0.05);與PTU組比較,L-T4組、聯(lián)合用藥組大鼠血清對氧磷酶-1含量顯著升高,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論甲狀腺功能減退可導(dǎo)致抗氧化應(yīng)激能力下降以及細胞凋亡的增加,左甲狀腺素鈉、兒茶素藥物治療可改善氧化應(yīng)激、減少細胞凋亡,兩藥聯(lián)合使用有協(xié)同作用。
[Abstract]:Objective to observe the effects of levothyroxine sodium, catechin and two drugs on serum 8-hydroxydeoxyguanosine (8-OHd G),) in rats with hypothyroidism. The effect on the content of phosphatase-1 and the content of apoptotic protein caspase-3 in hippocampal tissue. Methods Fifty adult male SD rats were randomly selected as control group. The remaining 40 rats were induced by propyl thiomitridine (PTU). The rats were divided into PTU group and catechin group. Levothyroxine sodium group (L-T4 group), levothyroxine sodium catechin group (combined medication group). The serum levels of 8-OHd G and p-oxonase-1 were measured by enzyme-linked immunosorbent assay (Elisa). The content of apoptotic protein caspase-3 in hippocampus of rats in each group was determined by Western blot. Results 1 compared with the control group, the content of serum 8-OHd G and the content of apoptotic protein caspase-3 in hippocampus in PTU group were significantly higher than those in control group (P0.05). Compared with PTU group, serum 8-OHd G content and hippocampal apoptotic protein caspase-3 content in L-T4 group and combined treatment group were significantly lower than those in control group (P0.05). The level of serum paraoxonase-1 in PTU group was significantly lower than that in control group (P0.05). Compared with PTU group, the level of serum paraoxonase-1 in L-T4 group and combined treatment group was significantly higher than that in L-T4 group (P0.05). Conclusion hypothyroidism can decrease the ability of anti-oxidative stress and increase the apoptosis of cells. The treatment of levothyroxine sodium and catechin can improve the oxidative stress and decrease the apoptosis of cells. The combination of the two drugs has synergistic effect.
