高濃度葡萄糖對(duì)C57小鼠視網(wǎng)膜神經(jīng)節(jié)細(xì)胞的視覺(jué)反應(yīng)特性的影響——一項(xiàng)離體生理研究

【摘要】:本研究旨在觀察視網(wǎng)膜神經(jīng)節(jié)細(xì)胞在高濃度葡萄糖下的視覺(jué)反應(yīng)特性.實(shí)驗(yàn)中,視杯平鋪于記錄腔,Ames緩沖液灌流,單細(xì)胞外記錄小鼠(Mus musculus)視網(wǎng)膜神經(jīng)節(jié)細(xì)胞.實(shí)驗(yàn)結(jié)果表明,高糖條件下,ON神經(jīng)節(jié)細(xì)胞的平均感受野大小(34.1±2.9,n=14)明顯小于OFF神經(jīng)節(jié)細(xì)胞的(49.3±0.3,n=12)(P0.0001).高滲條件下,可以觀察到類似的模式,即ON神經(jīng)節(jié)細(xì)胞的平均感受野大小小于OFF的(P0.0001).ON神經(jīng)節(jié)細(xì)胞的平均亮度閾值在高糖(P0.0001)或者高滲(P0.0002)條件下明顯升高.OFF神經(jīng)節(jié)細(xì)胞的平均亮度閾值在相同條件下(高糖:P0.01;高滲:P0.0002)也有升高.在高滲條件下,ON神經(jīng)節(jié)細(xì)胞的平均對(duì)比增益明顯低于OFF神經(jīng)節(jié)細(xì)胞的(P0.015).而在高糖條件下,ON神經(jīng)節(jié)細(xì)胞的平均增益明顯高于OFF神經(jīng)節(jié)細(xì)胞的(P0.0001).這些結(jié)果表明,高糖可縮小神經(jīng)節(jié)細(xì)胞的感受野,降低亮度敏感度,減弱對(duì)比增益.高糖對(duì)ON和OFF神經(jīng)節(jié)細(xì)胞的影響可能是通過(guò)不同的機(jī)制進(jìn)行的.
[Abstract]:The aim of this study was to observe the visual response of retinal ganglion cells at high glucose concentration. In the experiment, the (Mus musculus) retinal ganglion cells of mice were recorded in the visual cup, the Ames buffer was perfused with the recording chamber, and the (Mus musculus) retinal ganglion cells were recorded outside the single cell. The results showed that the average receptive field size of ON ganglion cells (34.1 鹵2.9) was significantly lower than that of OFF ganglion cells (49.3 鹵0.3) (P0.0001). Similar patterns can be observed under hypertonic conditions. That is, the average receptive field size of ON ganglion cells was smaller than that of OFF (P0.0001). ON ganglion cells), the mean luminance threshold of). ON ganglion cells increased significantly under high glucose (P0.0001) or hyperosmotic (P0.0002) conditions. The mean luminance threshold was in the same condition (high sugar: P0.01; Hyperosmotic: P0.0002) also increased. Under hypertonic condition, the average contrast gain of ON ganglion cells was significantly lower than that of OFF ganglion cells (P0.015). At high glucose, the average gain of ON ganglion cells was significantly higher than that of OFF ganglion cells (P0.0001). These results suggest that high glucose can reduce the receptive field, brightness sensitivity and contrast gain of ganglion cells. The effects of high glucose on ON and OFF ganglion cells may be mediated by different mechanisms.
