新疆地區(qū)漢族人群轉(zhuǎn)錄因子7類似物2基因多態(tài)性與2型糖尿病的相關(guān)性研究

【摘要】:目的:探討新疆地區(qū)漢族人群轉(zhuǎn)錄因子7類似物2(TCF7L2)基因rs11196218. rs10749127、rs290487、rs290481多態(tài)性與2型糖尿病(T2DM)的相關(guān)性。方法:選取2013年11月一2014年2月新疆醫(yī)科大學(xué)第一附屬醫(yī)院收治的漢族T2DM患者164例為T2DM組,另選取同時(shí)期體檢的健康人群195例為對(duì)照組,測(cè)量并記錄身高、體質(zhì)量、腹圍、血壓,檢測(cè)血糖、糖化血紅蛋白(HbA1c)、胰島素、C肽、血脂、尿酸水平。提取外周DNA,檢測(cè)TCF7L2基因rs11196218、rs10749127、 rs290487、rs290481基因型及等位基因頻率。多因素Logistic回歸分析T2DM的影響因素。結(jié)果:對(duì)照組與T2DM組收縮壓(SBP)比較,差異無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);T2DM組患者舒張壓(DBP)、三酰甘油(TG)、總膽固醇(TC)、低密度脂蛋白膽固醇(LDL-C)、空腹血糖及HbA 1c較對(duì)照組升高,高密度脂蛋白膽固醇(HDL-C)較對(duì)照組降低(P0.05)。對(duì)照組與T2DM組rs11196218、rs290481基因型和等位基因頻率分布,差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05);兩組rs10749127 TT基因型分布,差異有統(tǒng)計(jì)學(xué)意義(P0.05),rs290487 CC基因型分布及等位基因頻率分布,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。rs10749127的CC、CT、TT基因型T2DM患者間空腹血糖及尿酸水平比較,差異有統(tǒng)計(jì)學(xué)意義(P0.05)。rs290487的CC、CT、TT基因型間各指標(biāo)比較,差異均無(wú)統(tǒng)計(jì)學(xué)意義(P0.05)。多因素Logistic回歸分析結(jié)果顯示:rs290487CC基因型、腰圍、體質(zhì)指數(shù)(BMI)、空腹血糖及HbA1c與T2DM有回歸關(guān)系(P0.05)。結(jié)論:TCF7L2基因rs10749127、rs290487與新疆地區(qū)漢族人群的T2DM的發(fā)病率有關(guān),未發(fā)現(xiàn)rs11196218、rs290481與T2DM發(fā)病的關(guān)聯(lián)。
[Abstract]:Objective: to study the rs11196218. of transcription factor 7 analogue 2 (TCF7L2) gene in Xinjiang Han population. Association of rs10749127,rs290487,rs290481 polymorphism with type 2 diabetes mellitus (T2DM). Methods: from November 2013 to February 2014, 164 T2DM patients of Han nationality admitted in the first affiliated Hospital of Xinjiang Medical University were selected as T2DM group, and 195 healthy subjects were selected as control group. Height, body mass and abdominal circumference were measured and recorded. Blood pressure, blood glucose, glycosylated hemoglobin (HbA1c), insulin, C-peptide, blood lipids, uric acid levels. Peripheral DNA, was extracted to detect the rs11196218,rs10749127, rs290487,rs290481 genotype and allele frequency of TCF7L2 gene. Multivariate Logistic regression analysis was used to analyze the influencing factors of T2DM. Results: there was no significant difference in systolic blood pressure (SBP) between control group and T2DM group (P0.05). The diastolic blood pressure of (DBP), triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) fasting blood glucose and HbA 1c in T2DM group were higher than those in control group. High density lipoprotein cholesterol (HDL-C) was lower than that of control group (P0.05). The frequency distribution of rs11196218,rs290481 genotype and allele in control group and T2DM group were not significantly different (P0.05). The genotype distribution of rs10749127 TT in the two groups was statistically significant (P0.05), and the distribution of rs290487 CC genotype and allele frequency was statistically significant (P0.05). The CC,CT, of rs10749127 was statistically significant (P0.05). There were significant differences in fasting blood glucose and uric acid levels among T2DM patients with TT genotype (P0.05). There was no significant difference in CC,CT,TT genotypes of rs290487 (P0.05). Multivariate Logistic regression analysis showed that rs290487CC genotype, waist circumference, body mass index (BMI),) fasting blood glucose and HbA1c were correlated with T2DM (P0.05). Conclusion: TCF7L2 gene rs10749127,rs290487 is related to the incidence of T2DM in Xinjiang Han population, and no association between rs11196218,rs290481 and T2DM is found.
