2型糖尿病人外周血TIM4蛋白水平增高且與IL-1β負(fù)相關(guān)

【摘要】:目的探討2型糖尿病人(T2DM)中TIM4的蛋白表達(dá)水平及與炎性小體活化指標(biāo)IL-1β的相關(guān)性。方法選取新診斷T2DM患者作為病例組,正常體檢者為對(duì)照,利用流式細(xì)胞術(shù)檢測(cè)病人外周血單核細(xì)胞上TIM4的蛋白表達(dá)水平,同時(shí)用ELISA檢測(cè)血清中可溶性TIM4的水平,同時(shí)分析TIM4與IL-1β的相關(guān)性。結(jié)果與正常對(duì)照組相比,T2DM病人血清中可溶性TIM4明顯升高(P0.001),單核細(xì)胞上TIM4表達(dá)上調(diào)(P=0.01),TIM4與IL-1β亦呈負(fù)相關(guān)性趨勢(shì)(P=0.003 6,r=-0.404 5);與T2DM病人治療前相比,治療后外周血單核細(xì)胞上TIM4表達(dá)下降(P=0.002)。結(jié)論 T2DM病人血液中TIM4蛋白水平增高,且與IL-1β呈負(fù)相關(guān)性,提示TIM4作為負(fù)性調(diào)控分子參與T2DM的疾病進(jìn)程。
[Abstract]:Objective to investigate the expression of TIM4 protein in type 2 diabetes mellitus (T2DM) and its correlation with inflammatory corpuscular activation index (IL-1 尾). Methods the expression of TIM4 protein in peripheral blood monocytes and soluble TIM4 in serum were detected by flow cytometry in newly diagnosed T2DM patients and normal controls. The correlation between TIM4 and IL-1 尾 was also analyzed. Results compared with the normal control group, the serum soluble TIM4 in T2DM patients was significantly increased (P0.001), the expression of TIM4 in monocytes was up-regulated (P0. 01), and there was a negative correlation between TIM4 and IL-1 尾 (P0. 0036 r-0.404 5). The expression of TIM4 in peripheral blood monocytes was decreased after treatment compared with that in patients with T2DM (P0. 002). Conclusion the level of TIM4 protein in the blood of patients with T2DM is increased and negatively correlated with IL-1 尾, suggesting that TIM4 is a negative regulatory molecule involved in the disease progression of T2DM.
