DTBNP、DTDP促進(jìn)INS-1細(xì)胞葡萄糖刺激胰島素分泌

【摘要】:目的:探討巰基氧化還原試劑對葡萄糖刺激胰島素分泌(glucose-stimulated insulin secretion,GSIS)影響,進(jìn)而揭示其調(diào)節(jié)胰島素分泌的可能機(jī)制。方法 :INS-l細(xì)胞經(jīng)傳代培養(yǎng)3~4 d后在KRBH液中,37℃培養(yǎng)箱孵育30 min,再用含有不同濃度葡萄糖和巰基氧化還原試劑的KRBH液中培養(yǎng)60 min。然后留取上清液進(jìn)行胰島素測定。結(jié)果:(1)INS-1細(xì)胞在2.5、5、10、15、20 mmol/L葡萄糖濃度范圍內(nèi)胰島素分泌量逐漸增加,G5、G10、G15組間兩兩相比均有統(tǒng)計(jì)學(xué)意義(P0.05);(2)與G10組相比,G10+DTBNP、G10+DTDP組胰島素分泌量顯著增加(P0.05),且該效應(yīng)可以被DTT所消除。(3)DTBNP、DTDP均能增加NIF處理組胰島素分泌,但其增加幅度低于非NIF處理組(P0.05);(4)與非DIA組相比,G10+DIA+DTBNP、G10+DIA+DTDP組胰島素分泌增加幅度顯著減低(P0.05);(5)同G10組比較,G10+DIA+NIF+DTBNP、G10+DIA+NIF+DTDP組胰島素分泌值增加(P0.05)。結(jié)論 :本研究顯示巰基氧化還原試劑對GSIS產(chǎn)生調(diào)節(jié)作用。DTDP、DTBNP可能通過對K_ATP、L型Ca_V通道及IP_3受體活性的調(diào)節(jié),促進(jìn)胰島素分泌。
