CIDEC的亞細(xì)胞定位及其功能初步研究

【摘要】:CIDEC(也稱FSP27)高表達(dá)于脂肪組織,可以促進(jìn)細(xì)胞內(nèi)脂肪積累等。為探究CIDEC促進(jìn)脂滴融合的機(jī)制,本研究利用脂肪酸處理HepG2細(xì)胞,測(cè)定細(xì)胞內(nèi)脂滴直徑和數(shù)目的變化,發(fā)現(xiàn)處理后脂滴的直徑和數(shù)量無顯著性差異。將含有CIDEC的重組載體轉(zhuǎn)染細(xì)胞,脂肪酸處理24 h后測(cè)定脂滴直徑和數(shù)量變化,進(jìn)一步利用共聚焦顯微鏡分析其亞細(xì)胞定位,并檢測(cè)與脂滴生成、生長(zhǎng)等相關(guān)基因表達(dá)量的變化。結(jié)果表明,過表達(dá)CIDEC后脂滴的直徑顯著增加,脂滴的數(shù)量極顯著減少;亞細(xì)胞定位發(fā)現(xiàn),CIDEC位于脂滴周圍。此外,PLIN1、CREB1、CREB8的表達(dá)量有所下降,CIDEA、CFD表達(dá)量有所上升,推測(cè)CIDEC可能與這些蛋白互作引起脂滴融合。本研究結(jié)果為探究CIDEC促進(jìn)脂滴融合的分子機(jī)制奠定了基礎(chǔ)。
[Abstract]:CIDEC (also known as FSP27) is highly expressed in adipose tissue and can promote intracellular fat accumulation. In order to explore the mechanism of lipid droplet fusion promoted by CIDEC, the changes of lipid droplet diameter and number in HepG2 cells treated with fatty acids were measured. It was found that there was no significant difference in the diameter and number of lipid droplets after treatment. The recombinant vector containing CIDEC was transfected into the cells. After 24 h of fatty acid treatment, the diameter and quantity of lipid droplets were measured, and the subcellular localization was analyzed by confocal microscope, and the changes of gene expression related to lipid droplet formation and growth were detected. The results showed that the diameter of lipid droplets increased significantly and the number of lipid droplets decreased significantly after overexpression of CIDEC. In addition, the expression of CREB1 / CREB8 decreased and the expression of CIDEAA increased, which suggested that the interaction of CIDEC with these proteins might lead to lipid droplet fusion. The results laid a foundation for exploring the molecular mechanism of lipid droplet fusion promoted by CIDEC.
