【摘要】：目的探討不同強(qiáng)度運(yùn)動(dòng)處方對老年肥胖大鼠內(nèi)臟脂肪組織特異性絲氨酸蛋白酶抑制劑(Vaspin)mRNA、蛋白表達(dá)和血Vaspin濃度的影響。方法選取雄性清潔級SD幼鼠,按照生長發(fā)育期、壯年期和老年期三個(gè)階段增肥,建立老年肥胖大鼠模型,選取6只自然生長老年大鼠為空白對照組,選取18只肥胖老年大鼠為實(shí)驗(yàn)組,隨機(jī)分為3組,每組6只,分別為肥胖對照組、低強(qiáng)度運(yùn)動(dòng)組、中強(qiáng)度運(yùn)動(dòng)組。運(yùn)動(dòng)處方分別為12 m/min×15 min、4組/次、組間休息5 min、60 min/次;15 m/min×15 min、4組/次、組間休息5 min、60 min/次。跑臺坡度均為0°,5次/周,連續(xù)8周。運(yùn)動(dòng)干預(yù)后測內(nèi)臟脂肪組織Vaspin mRNA表達(dá)采用qRT-PCR、測蛋白表達(dá)采用WB、測血Vaspin濃度采用ELISA。結(jié)果運(yùn)動(dòng)干預(yù)后,(1)Vaspin mRNA表達(dá)(×10~3):肥胖對照組(25.761±11.581)高于空白對照組(12.161±5.648)(P0.01),低強(qiáng)度運(yùn)動(dòng)組(10.482±2.668)、中強(qiáng)度運(yùn)動(dòng)組(13.478±5.115)低于肥胖對照組(P0.01),中強(qiáng)度運(yùn)動(dòng)組高于低強(qiáng)度運(yùn)動(dòng)組,但差異無統(tǒng)計(jì)學(xué)意義(P0.05)。(2)Vaspin蛋白表達(dá):肥胖對照組高于空白對照組(P0.01),低強(qiáng)度運(yùn)動(dòng)組低于肥胖對照組(P0.05),但中強(qiáng)度運(yùn)動(dòng)組高于肥胖對照組(P0.01)。(3)血Vaspin濃度:肥胖對照組[(25.000±3.767)ng/m L]高于空白對照組[(23.765±3.045)ng/m L],低強(qiáng)度運(yùn)動(dòng)組[(25.637±5.240)ng/mL]和中強(qiáng)度運(yùn)動(dòng)組[(26.422±3.4175)ng/mL]高于肥胖對照組,但差異均無統(tǒng)計(jì)學(xué)意義(P0.05)。(4)血糖、胰島素敏感性指數(shù):肥胖對照組的血糖[(17.930±6.094)mg/dL]高于空白對照組[(11.636±3.568)mg/dL](P0.01),胰島素敏感性指數(shù)肥胖對照組(0.369±0.017)低于空白對照組(0.394±0.019),但差異無統(tǒng)計(jì)學(xué)意義(P0.05);低[(13.362±1.819)mg/dL]、中強(qiáng)度運(yùn)動(dòng)組[(12.842±1.905)mg/dL]血糖低于肥胖對照組(P0.05),而胰島素敏感性指數(shù)[低(0.385±0.018),中(0.396±0.015)]高于肥胖對照組(P0.01)。結(jié)論老年肥胖大鼠內(nèi)臟脂肪的Vaspin mRNA表達(dá)、蛋白表達(dá)和血漿含量偏高。低強(qiáng)度運(yùn)動(dòng)可降低Vaspin mRNA基因表達(dá)量和蛋白表達(dá),運(yùn)動(dòng)強(qiáng)度增加又提高Vaspin mRNA表達(dá)和蛋白表達(dá)以及血漿Vaspin水平。
[Abstract]:Objective to investigate the effects of different intensity exercise prescriptions on the expression of (Vaspin) mRNA, protein and the concentration of serum Vaspin in visceral adipose tissue specific serine protease inhibitor (Vaspin) mRNA,) in aged obese rats. Methods male SD young rats of clean grade were selected to gain weight according to the three stages of growth and development, adult and old. Six naturally growing aged rats were selected as the blank control group. Eighteen obese aged rats were randomly divided into 3 groups: obese control group, low intensity exercise group and moderate intensity exercise group. The exercise prescriptions were 12 m/min 脳 15 min for 4 groups, 5 minutes for 60 min/, 15 m/min 脳 15 minutes for each time, and 5 minutes for 60 min/. The slope of treadmill was 0 擄/ week for 8 weeks. After exercise intervention, the expression of Vaspin mRNA in visceral adipose tissue was measured by qRT-PCR, the expression of protein was measured by WB, and the concentration of blood Vaspin was measured by Elisa. Results after exercise intervention, (1) Vaspin mRNA expression in obese group (25.761 鹵11.581) was higher than that in blank control group (12.161 鹵5.648), low intensity exercise group (10.482 鹵2.668), moderate intensity exercise group (13.478 鹵5.115) was lower than obese control group (P0.01), and moderate intensity exercise group was higher than low intensity exercise group. But there was no significant difference (P0.05). (2) Vaspin protein expression: obese control group was higher than blank control group (P0.01), low-intensity exercise group was lower than obese control group (P0.05), but moderate intensity exercise group was higher than obese control group (P0.01). (3) serum Vaspin concentration: obesity control group [(25.000 鹵3.767) ng/m L] was high. In the blank control group [(23.765 鹵3.045) ng/m L], the low intensity exercise group [(25.637 鹵5.240) ng/mL] and the moderate intensity exercise group [(26.422 鹵3.4175) ng/mL] were higher than those in the obese control group. But there was no significant difference in blood glucose (P0.05). (4). Insulin sensitivity index (ISI) in obese group [(17.930 鹵6.094) mg/dL] was higher than that in blank control group [(11.636 鹵3.568) mg/dL] (P0.01), insulin sensitivity index in obese control group (0.369 鹵0.017) was lower than that in blank control group (0.394 鹵0.019), but the difference was not statistically significant (P0.05), low [(13.362 鹵1.819) mg/dL], moderate exercise group [(12.842 鹵1.905) mg/dL] In the obese control group (P0.05), the insulin sensitivity index [(0.385 鹵0.018), (0.396 鹵0.015)] was higher than that in the obese control group (P0.01). Conclusion the Vaspin mRNA expression, protein expression and plasma content of visceral fat were higher in aged obese rats. Low intensity exercise decreased the expression of Vaspin mRNA gene and protein, increased exercise intensity and increased the expression of Vaspin mRNA and protein, as well as the level of plasma Vaspin.
