不同強度運動處方對老年肥胖大鼠內(nèi)臟脂肪Vaspin mRNA、蛋白表達和血Vaspin濃度的影響
[Abstract]:Objective to investigate the effects of different intensity exercise prescriptions on the expression of (Vaspin) mRNA, protein and the concentration of serum Vaspin in visceral adipose tissue specific serine protease inhibitor (Vaspin) mRNA,) in aged obese rats. Methods male SD young rats of clean grade were selected to gain weight according to the three stages of growth and development, adult and old. Six naturally growing aged rats were selected as the blank control group. Eighteen obese aged rats were randomly divided into 3 groups: obese control group, low intensity exercise group and moderate intensity exercise group. The exercise prescriptions were 12 m/min 脳 15 min for 4 groups, 5 minutes for 60 min/, 15 m/min 脳 15 minutes for each time, and 5 minutes for 60 min/. The slope of treadmill was 0 擄/ week for 8 weeks. After exercise intervention, the expression of Vaspin mRNA in visceral adipose tissue was measured by qRT-PCR, the expression of protein was measured by WB, and the concentration of blood Vaspin was measured by Elisa. Results after exercise intervention, (1) Vaspin mRNA expression in obese group (25.761 鹵11.581) was higher than that in blank control group (12.161 鹵5.648), low intensity exercise group (10.482 鹵2.668), moderate intensity exercise group (13.478 鹵5.115) was lower than obese control group (P0.01), and moderate intensity exercise group was higher than low intensity exercise group. But there was no significant difference (P0.05). (2) Vaspin protein expression: obese control group was higher than blank control group (P0.01), low-intensity exercise group was lower than obese control group (P0.05), but moderate intensity exercise group was higher than obese control group (P0.01). (3) serum Vaspin concentration: obesity control group [(25.000 鹵3.767) ng/m L] was high. In the blank control group [(23.765 鹵3.045) ng/m L], the low intensity exercise group [(25.637 鹵5.240) ng/mL] and the moderate intensity exercise group [(26.422 鹵3.4175) ng/mL] were higher than those in the obese control group. But there was no significant difference in blood glucose (P0.05). (4). Insulin sensitivity index (ISI) in obese group [(17.930 鹵6.094) mg/dL] was higher than that in blank control group [(11.636 鹵3.568) mg/dL] (P0.01), insulin sensitivity index in obese control group (0.369 鹵0.017) was lower than that in blank control group (0.394 鹵0.019), but the difference was not statistically significant (P0.05), low [(13.362 鹵1.819) mg/dL], moderate exercise group [(12.842 鹵1.905) mg/dL] In the obese control group (P0.05), the insulin sensitivity index [(0.385 鹵0.018), (0.396 鹵0.015)] was higher than that in the obese control group (P0.01). Conclusion the Vaspin mRNA expression, protein expression and plasma content of visceral fat were higher in aged obese rats. Low intensity exercise decreased the expression of Vaspin mRNA gene and protein, increased exercise intensity and increased the expression of Vaspin mRNA and protein, as well as the level of plasma Vaspin.
