高原低氧條件下大鼠鐵代謝及其相關(guān)因子表達(dá)的變化

【摘要】:目的探討高原低氧條件下大鼠鐵代謝及其相關(guān)因子表達(dá)變化。方法選取雄性SD大鼠50只,將大鼠分別在低氧環(huán)境[低壓氧艙(模擬海拔5 000 m)飼養(yǎng)]下飼養(yǎng)7、15、30 d(低氧7 d、15 d、30 d組),常氧條件下[西寧地區(qū)(海拔2 260 m)飼養(yǎng)]飼養(yǎng)7、15、30 d(常氧7 d、15 d、30 d組)。分別于飼養(yǎng)7、15、30 d測(cè)定大鼠外周血血常規(guī)、血清鐵(SI)、血清鐵蛋白(SF)水平,qRT-PCR和Western blotting法測(cè)大鼠肝組織鐵調(diào)素(Hepc)、膜結(jié)合型鐵調(diào)素調(diào)節(jié)蛋白(m-HJV)、Ⅱ型跨膜絲氨酸蛋白酶(TMPRSS6)及肌肉組織可溶型鐵調(diào)素調(diào)節(jié)蛋白(s-HJV)mRNA和蛋白表達(dá)量,qRT-PCR和ELISA法測(cè)大鼠肝臟Hepc mRNA和蛋白表達(dá)量。結(jié)果低氧7 d、15 d、30 d組大鼠血紅蛋白(Hb)、紅細(xì)胞壓積(HCT)、SF水平均高于常氧同時(shí)間組(P均0.01),低氧30 d組SI水平高于常氧同時(shí)間組(P0.05),隨著缺氧時(shí)間延長(zhǎng)Hb、HCT、SI、SF水平逐漸升高(P均0.05)。低氧15 d、30 d組肝組織Hepc mRNA及蛋白表達(dá)量均低于常氧同時(shí)間組(P0.01或0.05),且隨著缺氧時(shí)間延長(zhǎng)其表達(dá)水平逐漸降低(P均0.01)。低氧各組間組大鼠肝組織m-HJV mRNA及蛋白表達(dá)量均低于常氧同時(shí)間組(P均0.01),隨缺氧時(shí)間延長(zhǎng)m-HJV逐漸降低(P均0.01)。低氧各時(shí)間組大鼠肌肉組織s-HJV mRNA及蛋白表達(dá)量、肝組織TMPRSS6 mRNA及蛋白表達(dá)量均高于常氧同時(shí)間組(P均0.01),且隨著缺氧時(shí)間延長(zhǎng)表達(dá)量逐漸升高(P均0.01)。結(jié)論高原低氧條件下,大鼠肝組織TMPRSS6表達(dá)量升高,可能裂解m-HJV成為s-HJV,使Hepc轉(zhuǎn)錄受抑制,最終導(dǎo)致SI水平升高。
