【摘要】：胰島素抵抗、肥胖病和2型糖尿病與慢性炎癥有著密切關(guān)系,慢性炎癥中促炎癥細(xì)胞因子如腫瘤壞死因子-α(tumor necrosis-α,TNF-α)、白介素-6(interleukin-6,IL-6)等通過(guò)激活經(jīng)典的IKKα,IKKβ途徑和非經(jīng)典的IKKε,TDK1途徑,激活NF-кB途徑,引起胰島素抵抗、肥胖病和2型糖尿病,其中IKKβ在經(jīng)典的IKKα和IKKβ途徑中起著主導(dǎo)作用。阻斷IKKα和IKKβ途徑,可顯著減少炎癥、增加胰島素敏感性,降低體重和減少2型糖尿病。因此,阻斷IKKα和IKKβ途徑,可作為研究治療胰島素抵抗、肥胖病和2型糖尿病的一個(gè)新的作用靶點(diǎn)。
[Abstract]:Insulin resistance, obesity and type 2 diabetes are closely related to chronic inflammation. In chronic inflammation, cytokines such as tumor necrosis-alpha (TNF- 偽), interleukin-6 (IL-6) activate the classical IKK 偽 -IKK 尾 pathway and the non-classical IKK 蔚 TDK1 pathway. NF-B pathway is activated to induce insulin resistance, obesity and type 2 diabetes, in which IKK 尾 plays a leading role in the classical IKK 偽 and IKK 尾 pathways. Blocking the IKK 偽 and IKK 尾 pathway significantly reduced inflammation, increased insulin sensitivity, reduced body weight and reduced type 2 diabetes. Therefore, blocking the IKK 偽 and IKK 尾 pathway may be a new target for the study of insulin resistance, obesity and type 2 diabetes.
