肥胖大鼠體內(nèi)鈣代謝狀態(tài)的變化及鈣與肥胖關(guān)系的研究
[Abstract]:Aim: to study the changes of calcium metabolism in rats with obesity and the effect of calcium supplementation on obesity so as to provide a theoretical basis for the further study of the relationship between obesity and calcium. Methods: forty male Wistar rats were randomly divided into control group (n = 10) and obese group (n = 30). After 8 weeks, 10 rats in the control group and 10 rats in the obese group were killed. Serum calcium, alkaline phosphatase, parathyroid hormone, calcitonin, vitamin D receptor, calcium binding protein 25 hydroxyvitamin D. The expression of vitamin D receptor and calcium binding protein gene was detected by real-time quantitative PCR in small intestine. The second part of the experiment was continued with the remaining 20 obese rats. Twenty obese rats were randomly divided into obese control group (n = 10) and calcium supplement group (n = 10). At the same time, the obese control group was given the same amount of normal saline for 15 days. The serum samples were collected to detect the following indexes: blood lipids four items, osteocalcin 25 hydroxyvitamin D. The expression of leptin and fatty acid synthase gene was detected by real-time quantitative PCR in subcutaneous white adipose tissue. Results compared with the control group, the serum calcium level and alkaline phosphatase activity of obese rats were lower, and the differences were statistically significant (P0.05) .2.Compared with the control group, the serum parathyroid hormone levels of obese rats were higher. Calcitonin level was lower, and the difference was statistically significant (P0.05), the serum calcium binding protein level was lower, the vitamin D receptor level was higher and the vitamin D level was lower in obesity group. Compared with the control group, the expression of calcium binding protein gene and vitamin D receptor gene in the small intestine of obese rats were lower than those of the control group. The differences were statistically significant (P0.05) .4Compared with the obese control group, the body weight index of rats in calcium supplementation group was lower, and the difference was statistically significant (P0.05), and the total cholesterol, triglyceride and triglyceride in calcium supplementation group were significantly lower than those in the obese control group (P0.05). The low density lipoprotein level was lower, and the difference was statistically significant (P0.05), the high density lipoprotein level was higher, and the difference was statistically significant (P0.05). The serum levels of osteocalcin 25 hydroxyvitamin D in the calcium supplementation group were higher, and the differences were statistically significant (P0.05) .6. Compared with the obese control group, the leptin gene in the white adipose tissue in the calcium supplementation group was significantly higher than that in the control group. Fatty acid synthase gene expression was low, and the differences were statistically significant (P0.05). Conclusion: weight 1, obesity can decrease serum calcium level, increase alkaline phosphatase activity, increase serum parathyroid hormone level, calcitonin level, calcium binding protein level, and increase serum parathyroid hormone level, calcitonin level and calcium binding protein level. The level of vitamin D receptor decreased. 3. Obesity decreased the expression of calcium binding protein gene and vitamin D receptor gene in the small intestine of rats, and calcium supplementation decreased the Lees index of rat body weight. The levels of total cholesterol, triglyceride, low density lipoprotein, high density lipoprotein, calcium supplementation increased the serum osteocalcin 25 hydroxyl vitamin D level, and calcium supplementation increased the leptin gene in white adipose tissue of rats. The expression of fatty acid synthase gene decreased.
