高尿酸血癥細胞模型的構建及其在降尿酸肽篩選中的應用

【摘要】:高尿酸血癥是由于嘌呤代謝紊亂使尿酸生成增加和(或)排泄減少所致的代謝性疾病,是引起痛風的重要生化基礎。本文建立了一種高尿酸血癥肝細胞模型,并研究了乳清蛋白肽的降尿酸活性。具體步驟為:將肝細胞鋪板,分為空白對照組和模型組,48 h后,模型組給予一定濃度的腺苷溶液,空白對照組給予基礎培養(yǎng)基,36 h后,加入黃嘌呤氧化酶繼續(xù)孵育12 h,收集細胞上清液,高效液相測定尿酸含量。結果顯示模型組尿酸含量顯著高于空白對照組,且腺苷的最佳誘導濃度、細胞鋪板密度及誘導時間分別為2.5 mmol/L、10~5個/m L、36 h。采用最佳建模條件,將非布索坦和乳清蛋白肽用于該模型,結果顯示相對于模型組,非布索坦組尿酸含量顯著降低,并呈一定的濃度劑量效應,該結果進一步表明高尿酸血癥模型構建成功。采用乳清蛋白酶解液孵育肝細胞后,當乳清蛋白肽達到一定濃度時,相對于模型組其尿酸生成量顯著降低,表明乳清蛋白酶解物具有一定的降尿酸活性。
[Abstract]:Hyperuricemia is a metabolic disease caused by increased uric acid production and / or decreased excretion due to purine metabolic disorder, which is an important biochemical basis for gout. A hepatocyte model of hyperuricemia was established and the antiuric acid activity of whey protein peptide was studied. The specific steps were as follows: the hepatocytes were divided into blank control group and model group after 48 hours, the model group was given a certain concentration of adenosine solution, and the blank control group was given basic culture medium for 36 h. Xanthine oxidase was added for 12 hours to collect cell supernatant and determine uric acid content in high performance liquid phase. The results showed that the uric acid content in the model group was significantly higher than that in the control group, and the optimal concentration of adenosine, the cell paving density and the induction time were 2.5 mmol 路L ~ (-1) ~ (10 ~ 5) / mL ~ (-1) for 36 h, respectively. The results showed that the uric acid content of non-buzotam and whey protein peptide groups was significantly decreased compared with the model group, and the concentration of uric acid in the non-buzotam group was in a dose-dependent manner. The results further indicated that the model of hyperuricemia was successfully constructed. After incubating hepatocytes with whey proteolysis solution, when the concentration of whey protein peptide reached a certain concentration, the uric acid production was significantly decreased compared with the model group, which indicated that whey protease hydrolysate had a certain uric acid reducing activity.
