胰高血糖素樣肽-1對(duì)AGEs誘導(dǎo)H9C2心肌細(xì)胞自噬的影響

【摘要】:目的觀察胰高血糖素樣肽-1(GLP-1)對(duì)晚期糖基化終產(chǎn)物(AGEs)誘導(dǎo)H9C2心肌細(xì)胞自噬(Autophagy)的影響,初步探討GLP-1心肌細(xì)胞保護(hù)作用與自噬活性關(guān)系。方法將傳代培養(yǎng)H9C2心肌細(xì)胞隨機(jī)分4組:(1)Control組:加入0.9%生理鹽水;(2)AGEs組:加入100 mg·L~(-1)AGEs;(3)AGEs+GLP-1組:同時(shí)加入100 mg·L~(-1)AGEs和10 nmol·L~(-1)GLP-1;(4)AGEs+GLP-1+Rapamycin組:同時(shí)加入100 mg·L~(-1)AGEs、10 nmol·L~(-1)GLP-1及5μmol·L~(-1)Rapamycin(自噬誘導(dǎo)劑)。各組在經(jīng)上述預(yù)處理24 h后,分別用CCK-8法檢測(cè)細(xì)胞存活率Hoechst 33258試劑盒檢測(cè)細(xì)胞的凋亡率,DCFH-DA熒光探針檢測(cè)細(xì)胞內(nèi)活性氧(ROS)水平,流式細(xì)胞術(shù)檢測(cè)細(xì)胞自噬溶酶體形成,Western blot法檢測(cè)自噬相關(guān)蛋白(LC3Ⅱ,Beclin-1)表達(dá)。結(jié)果 (1)與Control組相比,AGEs組細(xì)胞生存率明顯減少,細(xì)胞內(nèi)ROS水平明顯增高,細(xì)胞自噬溶酶體水平及自噬相關(guān)蛋白(LC3Ⅱ、Beclin-1)表達(dá)均明顯增加;(2)與AGEs組相比,AGEs+GLP-1組細(xì)胞生存率明顯增高,細(xì)胞內(nèi)ROS水平、細(xì)胞自噬溶酶體水平及自噬相關(guān)蛋白(LC3Ⅱ、Beclin-1)表達(dá)均明顯下降;(3)與AGEs組相比,AGEs+GLP-1+rapamycin組中細(xì)胞內(nèi)ROS水平降低,而細(xì)胞生存率、細(xì)胞自噬溶酶體水平及自噬相關(guān)蛋白(LC3Ⅱ、Beclin-1)表達(dá)未見差異。結(jié)論 (1)AGEs可導(dǎo)致H9C2心肌細(xì)胞內(nèi)ROS升高,誘導(dǎo)細(xì)胞損傷并激活細(xì)胞自噬;(2)GLP-1對(duì)AGEs誘導(dǎo)的H9C2心肌細(xì)胞損傷具有保護(hù)作用,其保護(hù)機(jī)制可能與抑制細(xì)胞自噬活性有關(guān)。
[Abstract]:Objective to observe the effect of glucagon like peptide 1 (GLP-1) on autophagy (Autophagy) induced by advanced glycation end products (ages), and to explore the relationship between the protective effect of GLP-1 and autophagy activity. Methods cultured H9C2 cardiomyocytes were randomly divided into four groups: (1) Control group: 0. 9% saline; (2) ages group: 100 mg L ~ (-1) ages; (3) ages GLP-1 group: 100 mg L ~ (-1) ages and 10 nmol L ~ (-1) GLP-1; (4) ages GLP-1 Rapamycin group: 100 mg L ~ (-1) ages 10 nmol L ~ (-1) GLP-1 and 5 渭 mol L ~ (-1) rapamycin.
