血管活性腸肽對實(shí)驗(yàn)性自身免疫性腦脊髓炎大鼠腦組織IFN-γ、IL-4因子含量的影響

【摘要】:目的:探討血管活性腸肽(VIP)對實(shí)驗(yàn)性自身免疫性腦脊髓炎(EAE)大鼠腦組織IFN-γ、IL-4含量的影響。方法:將60只健康雌性Wistar大鼠隨機(jī)分成正常對照組,EAE對照組,VIP低劑量、高劑量干預(yù)組。利用髓鞘堿性蛋白(MBP)+完全福氏佐劑(CFA)建立EAE模型。自造模當(dāng)日起,每隔1 d分別對VIP低、高劑量干預(yù)組大鼠腹腔注射VIP 4 nmol·kg-1(0.2 ml)、16 nmol·kg-1(0.8 ml),正常對照組及EAE對照組則給予生理鹽水0.8 ml,連續(xù)10 d。觀察大鼠發(fā)病情況、腦組織中IFN-γ、IL-4因子含量及腦組織HE染色。結(jié)果:與EAE對照組比較,VIP各劑量干預(yù)組大鼠發(fā)病潛伏期延長、進(jìn)展期時間縮短、發(fā)病高峰期神經(jīng)功能障礙評分(NDS)降低;腦組織中IFN-γ含量降低、IL-4含量升高,且VIP高劑量干預(yù)組效果最明顯;腦組織HE染色顯示炎癥細(xì)胞浸潤程度下降。結(jié)論:VIP通過降低腦組織中IFN-γ、升高IL-4含量減輕腦組織炎癥細(xì)胞浸潤,發(fā)揮對EAE的免疫調(diào)節(jié)作用。
[Abstract]:Aim: to investigate the effect of vasoactive intestinal peptide (VIP) on the content of IFN- 緯 -IL-4 in the brain of rats with experimental autoimmune encephalomyelitis (EAE). Methods: sixty healthy female Wistar rats were randomly divided into two groups: normal control group, low dose group and high dose group. The EAE model was established by using the complete Freund's adjuvant CFAs of myelin basic protein (MBP). From the same day, the rats in the low VIP group and the high dose group were injected intraperitoneally with VIP 4 nmol kg-1(0.2 Mel 16 nmol kg-1(0.8 ml, while the normal control group and EAE control group were given normal saline 0. 8 ml for 10 days. The incidence, the level of IFN- 緯 -IL-4 in brain tissue and HE staining in brain tissue were observed. Results: compared with the control group, the incubation period was prolonged, the time of progression was shortened, the neurologic dysfunction score was decreased, the content of IFN- 緯 in brain tissue was decreased, and the content of IL-4 was increased in the brain tissue. The effect of high dose VIP intervention group was the most obvious, and HE staining of brain tissue showed that the degree of inflammatory cell infiltration decreased. Conclusion by decreasing IFN- 緯 in brain tissue, increasing the content of IL-4 can reduce the infiltration of inflammatory cells in brain tissue and play an important role in the immune regulation of EAE.
