硫辛酸對(duì)糖尿病小鼠氧化還原狀態(tài)及大血管并發(fā)癥的影響

【摘要】:目的建立糖尿病小鼠模型,研究硫辛酸(LA)干預(yù)后氧化還原狀態(tài)、糖代謝酶活性和大血管的變化。方法小鼠分為正常對(duì)照(NC)組、糖尿病(DM)組和硫辛酸干預(yù)(LA)組,每組各10只,檢測(cè)FBG、FIns,谷胱甘肽還原型氧化型比值(GSH/GSSG),超氧化物歧化酶活性抑制率(SODI),肝臟丙酮酸羧化酶(PC)和己糖激酶(HK)活性;HE染色觀察胰島細(xì)胞、電鏡觀察腹主動(dòng)脈的微結(jié)構(gòu)。結(jié)果 LA組干預(yù)6周后,FBG低于DM組[(10.05±0.40)vs(7.03±0.26)mmol/L,P0.05)];干預(yù)結(jié)束后GSH/GSSG、肝臟PC活性低于DM組[(23.15×10~(-2)±7.99×10~(-2))vs(36.20×10~(-2)±1.14×10~(-2)),(2.87×10~(-2)±0.32×10~(-2))vs(17.30×10~(-2)±4.01×10~(-2))Abs/μg pro/min,P0.05];胰島細(xì)胞體積和排列、腹主動(dòng)脈內(nèi)皮細(xì)胞形態(tài)等較DM組恢復(fù)。結(jié)論 LA可以改善糖尿病小鼠模型氧化應(yīng)激狀態(tài),減輕胰島細(xì)胞和主動(dòng)脈內(nèi)皮的損傷。
[Abstract]:Objective to establish a diabetic mouse model and to study the redox state, the activity of glucose metabolism enzymes and the changes of large blood vessels after the intervention of lipoxylic acid (LAA). Methods the mice were divided into normal control group (n = 10), DM group (n = 10) and lipoic acid intervention group (n = 10). The activities of FBGN FIns, GSH / GSSGG, SOD activity inhibition rate (SODIN), hepatic pyruvate carboxylase (PCP) and hexokinase (HKK) were detected by HE staining, and the microstructures of abdominal aorta were observed by electron microscope. 緇撴灉 LA緇勫共棰,
