【摘要】:目的:通過檢測不同海拔同一時間下大鼠肺組織IL-6、IL-8及IL-10的水平,來進(jìn)一步探討高原低氧對氣道炎癥及組織損傷的機(jī)制。方法:選擇成年雄性Wistar大鼠40只,均購買于甘肅中醫(yī)學(xué)院,隨機(jī)分3組:對照組(n=8,蘭州,1500米)、中度海拔組(n=16,西寧,2260米)、模擬高海拔組(n=16,低壓氧艙,模擬海拔5000米),對照組大鼠于當(dāng)天就地取材,中度海拔組大鼠飼養(yǎng)于青海大學(xué)醫(yī)學(xué)院動物房,海拔2260m,按晝夜采光,通風(fēng)良好,室溫約25℃左右,定期進(jìn)行消毒,大鼠自由攝食及飲水。特高海拔組大鼠飼養(yǎng)于青海大學(xué)醫(yī)學(xué)院高原醫(yī)學(xué)研究中心的低壓氧艙,氧艙條件為:海拔5000m,壓力:52889Pa~53001Pa,氧分壓:42mm Hg,溫度:16.9℃-24℃,濕度:37.3%-51%。動物的攝食、飲水、清潔及消毒均在低壓氧艙內(nèi)進(jìn)行。中度海拔組和模擬高海拔組根據(jù)觀察時間不同分為1天、30天共4組,每組8只,本課題選取觀察時間為1天和30天的大鼠肺組織上清液,酶聯(lián)免疫吸附法檢測大鼠肺組織勻漿中IL-6、IL-8及IL-10的表達(dá)。結(jié)果:在中度海拔組:MA1組IL-6、IL-8、IL-10水平高于對照組(P0.05),其余各組與對照組無統(tǒng)計(jì)學(xué)差異(P0.05);在模擬高海拔組:其各組IL-6、IL-8水平均高于對照組及中度海拔組(P0.05);IL-10均低于對照組及中海拔組(P0.05)。結(jié)論:對照組剛進(jìn)入中度海拔環(huán)境會引起大鼠肺組織炎癥反應(yīng)增強(qiáng),同時抑炎作用也得以提升,表明中海拔環(huán)境對肺組織氣道炎癥反應(yīng)存在一定影響;高海拔低氧條件導(dǎo)致肺組織炎癥反應(yīng)加重而抑制炎癥反應(yīng)的能力減弱。
[Abstract]:Aim: to investigate the mechanism of airway inflammation and tissue damage induced by high altitude hypoxia by detecting the levels of IL-6 IL-8 and IL-10 in lung tissue of rats at the same time at different altitude. Methods: forty adult male Wistar rats were randomly divided into three groups: control group (n = 8), Lanzhou (1 500 m), moderate altitude (n = 16), Xining (2 260 m), simulated high altitude group (n = 16) and low pressure oxygen chamber (P < 0.01). The rats in the control group were collected locally on the same day, and the rats in the moderate altitude group were fed in the animal room of the College of Medicine of Qinghai University at 2260 m above sea level.
