【摘要】：目的探索骨硬化蛋白單克隆抗體(Scl-Ab)對去勢大鼠股骨干骨折愈合影響的實驗研究。方法 30只健康雌性SD大鼠隨機行假手術(Sham,n=5)和切除雙側卵巢(OVX,n=25)手術12w后每組各取5只大鼠處死取股骨行雙能X線儀檢測確保骨質(zhì)疏松的建立,隨后OVX組大鼠建立左側股骨干中段骨折模型隨機的分成2組:OVX組及骨硬化蛋白單克隆抗體治療(Scl-Ab)組。術后第1天Scl-Ab組開始給予藥物治療:Scl-Ab皮下注射(劑量25 mg/kg,每周2次)直至8 w,8 w時所有大鼠處死取股骨行Micro-CT、硬組織切片檢測及HE組織切片檢測。結果 Micro-CT結果顯示:Scl-Ab組和OVX組相比,骨折端有較多的新生骨形成,且有較高BV/TV、Tb.Th、Tb.N和較低的Tb.Sp值,比較有明顯的統(tǒng)計學意義。HE結果表明Scl-Ab組骨折端有更多的骨痂形成,且骨痂更加成熟。硬組織切片觀察結果表明:Scl-Ab組骨折端骨痂礦化速度更快,和OVX組表明有統(tǒng)計學意義。結論 Scl-Ab通過增加骨折端骨量及促進骨組織礦化來治療去卵巢大鼠股骨中段骨折的愈合。
[Abstract]:Objective to investigate the effect of bone sclerosing protein monoclonal antibody Scl-Abon on femoral shaft fracture healing in ovariectomized rats. Methods Thirty healthy female Sprague-Dawley rats were randomly divided into two groups: sham-operated rats (5) and bilateral ovariectomies (OVX) 25). After 12 weeks of operation, 5 rats in each group were killed and femurs were sacrificed to ensure the establishment of osteoporosis. Then the left femoral shaft fracture model was established in OVX group. The rats were randomly divided into 2 groups: 1% OVX group and 2 groups treated with bone sclerosing protein monoclonal antibody (OVX) group. On the first day after operation, Scl-Ab group was given subcutaneous injection of 25 mg / kg Scl-Ab (25 mg / kg, twice a week) until 8 weeks after 8 weeks of operation, all rats were killed to perform Micro-CTT, hard tissue sections and HE tissue sections. Results compared with OVX group, the Micro-CT results showed that there were more new bone formation, higher BV / TVTV Tb. Thal Tb.N and lower Tb.Sp value in Scl-Ab group than in OVX group. The results showed that there were more callus formation at the fracture end in Scl-Ab group. And the callus is more mature. The results of hard tissue sections showed that the callus mineralization rate of the fracture tip in the Scl-Ab group was faster than that in the OVX group. Conclusion Scl-Ab can increase the bone mass and promote the mineralization of bone tissue to treat the healing of middle femur fracture in ovariectomized rats.
