不同干預(yù)療法對去卵巢骨質(zhì)疏松大鼠干預(yù)作用的對比研究

【摘要】:實(shí)驗(yàn)?zāi)康模罕疚牟捎萌ヂ殉补琴|(zhì)疏松大鼠模型,對比觀察了全身垂直振動、跑臺運(yùn)動、金雀異黃酮、雌激素和氯化鋰對去卵巢骨質(zhì)疏松大鼠骨密度、骨微結(jié)構(gòu)、成骨調(diào)節(jié)相關(guān)蛋白表達(dá)的影響以及對子宮的副作用,旨在為絕經(jīng)后骨質(zhì)疏松的臨床防治提供實(shí)驗(yàn)參考依據(jù)。實(shí)驗(yàn)方法:將84只3月齡雌性SD大鼠按體重分層后隨機(jī)分為假手術(shù)組、去卵巢組、振動組、跑臺組、金雀異黃酮組、氯化鋰組和雌激素組,去卵巢第11周時,開始進(jìn)行不同干預(yù)處理。跑臺組每天進(jìn)行1次時間45min、速度18m/min、坡度5。的跑臺訓(xùn)練;振動組每天進(jìn)行2次15min、頻率90HZ、7次/周的振動治療;金雀異黃酮組每天按體重灌胃1次金雀異黃酮,劑量為1mg/kg體重;氯化鋰組每周按體重腹腔注射3次氯化鋰,劑量為15mg/kg;雌激素組每周按體重頸部皮下注射3次17β-雌二醇,劑量為25ug/kg.干預(yù)處理8周時,于末次處理結(jié)束36-48小時內(nèi),腹主動脈取血處死各組大鼠,用雙能臌骨密度儀檢測大鼠離體腰椎骨密度,用micro-CT檢測腰椎骨微結(jié)構(gòu),用721分光光度計(jì)檢測血清ALP水平,用ELISA檢測血清P-catenin水平,用NBT-BCIP染色法檢測全骨髓細(xì)胞ALP的變化,用Western blot方法檢測骨組織和子宮P-GSK-3β、ER-a和ERR-a蛋白表達(dá)的變化。實(shí)驗(yàn)結(jié)果: (1)雌激素組、金雀異黃酮組、跑臺組和振動組大鼠體重均顯著低于去卵巢組,而氯化鋰組差異無顯著性。(2)雌激素組和跑臺組血清E2水平顯著高于去卵巢組,而金雀異黃酮組、氯化鋰組和振動組差異無顯著性;雌激素組、金雀異黃酮組、跑臺組、振動組和氯化鋰組大鼠血清ALP水平均顯著低于去卵巢組;各組大鼠血清β-catenin水平差異均無顯著性。(3)跑臺組腰椎離體骨密度顯著高于去卵巢組,而雌激素素組、振動組、金雀異黃酮組和氯化鋰組差異均無顯著性。 (4)氯化鋰組大鼠體積骨密度、骨體積分?jǐn)?shù)和骨小梁數(shù)目顯著高于去卵巢組,骨小梁間隙、骨小梁厚度和結(jié)構(gòu)模型指數(shù)無顯著變化,而雌激素組、金雀異黃酮組、跑臺組和振動組所有檢測指標(biāo)均無顯著變化。 (5)雌激素組、振動組和氯化鋰組大鼠股骨和子宮P-GSK-3β表達(dá)水平均顯著高于去卵巢組,而骨髓細(xì)胞P-GSK-3β表達(dá)水平無顯著變化;金雀異黃酮組股骨和骨髓細(xì)胞P-GSK-3β表達(dá)水平顯著高于去卵巢組,而子宮P-GSK-3β表達(dá)水平差異無顯著性;跑臺組股骨、骨髓細(xì)胞和子宮P-GSK-3β表達(dá)水平均顯著高于去卵巢組;(6)雌激素組股骨ER-α表達(dá)水平均顯著高于去卵巢組,脛骨、第三腰椎、骨髓細(xì)胞和子宮ER-α表達(dá)水平均顯著低于去卵巢組;金雀異黃酮組股骨ER-a表達(dá)水平顯著高于去卵巢組,脛骨和第三腰椎ER-α表達(dá)水平顯著低于去卵巢組,而骨髓細(xì)胞和子宮ER-α表達(dá)水平差異無顯著性;氯化鋰組股骨ER-α表達(dá)水平顯著高于去卵巢組,脛骨、第三腰椎和子宮ER-α表達(dá)水平顯著低于去卵巢組,而骨髓細(xì)胞ER-α表達(dá)水平差異無顯著性;跑臺組股骨、第三腰椎和骨髓細(xì)胞ER-α表達(dá)水平顯著高于去卵巢組,而脛骨和子宮ER-α表達(dá)水平顯著低于去卵巢組;振動組大鼠股骨ER-α表達(dá)水平顯著高于去卵巢組,脛骨、第三腰椎和子宮ER-α表達(dá)水平顯著低于去卵巢組,而骨髓細(xì)胞ER-α表達(dá)水平差異無顯著性。 (7)雌激素組股骨、脛骨和子宮ERR-a蛋白表達(dá)水平顯著低于去卵巢組,而第三腰椎和骨髓細(xì)胞ERR-a蛋白表達(dá)水平顯著高于去卵巢組;金雀異黃酮組股骨和脛骨ERR-a蛋白表達(dá)水平顯著低于去卵巢組,第三腰椎、骨髓細(xì)胞和子宮ERR-a蛋白表達(dá)水平顯著高于去卵巢組;氯化鋰組股骨、脛骨和第三腰椎ERR-a蛋白表達(dá)水平顯著低于去卵巢組,骨髓細(xì)胞ERR-a表達(dá)水平顯著高于去卵巢組,而子宮ERR-a表達(dá)水平差異無顯著性;跑臺組大鼠股骨、脛骨和子宮ERR-a蛋白表達(dá)水平顯著低于去卵巢組,第三腰椎和骨髓細(xì)胞ERR-a蛋白表達(dá)水平顯著高于去卵巢組;振動組大鼠脛骨和子宮ERR-a蛋白表達(dá)水平顯著低于去卵巢組,第三腰椎和骨髓細(xì)胞ERR-a蛋白表達(dá)水平顯著高于去卵巢組,而股骨ERR-a蛋白表達(dá)水平差異無顯著性。實(shí)驗(yàn)結(jié)論:跑臺運(yùn)動能增加去卵巢骨質(zhì)疏松大鼠血清E2水平;跑臺運(yùn)動、振動、金雀異黃酮、氯化鋰和雌激素均能降低去卵巢骨質(zhì)疏松大鼠血清ALP水平;跑臺運(yùn)動能減緩去卵巢骨質(zhì)疏松大鼠骨密度的下降,而氯化鋰能改善去卵巢骨質(zhì)疏松大鼠的骨微結(jié)構(gòu):跑臺運(yùn)動、振動、金雀異黃酮、氯化鋰和雌激素均部位和組織依賴性地調(diào)節(jié)去卵巢骨質(zhì)疏松大鼠組織P-GSK-3β、ER-α和ERR-a的蛋白表達(dá)水平;跑臺運(yùn)動、振動、金雀異黃酮和氯化鋰均無雌激素樣增加去卵巢骨質(zhì)疏松大鼠子宮重量的副作用。
[Abstract]:Objective : To investigate the effects of whole body vertical vibration , treadmill exercise , genistein , estrogen and lithium chloride on the expression of bone mineral density , bone microstructure , bone - forming regulation - related protein in ovariectomized osteoporotic rats , and to provide experimental reference for the clinical prevention and treatment of postmenopausal osteoporosis .
The vibration group was treated twice a day with a frequency of 90HZ and 7 times per week .
The genistein group was given 1 dose of genistein per day , and the dosage was 1 mg / kg body weight ;
The levels of ALP in isolated lumbar vertebrae were measured by means of micro - CT . The levels of serum ALP were measured by means of micro - CT . The levels of serum ALP were detected by using the method of Western blot . The serum levels of serum E2 in estrogen group , genistein group , treadmill group and vibrating group were significantly higher than those in ovariectomized group , while the difference between estrogen group and serum E2 level was higher than that in ovariectomized group , while the difference between the group of genistein and lithium chloride was not significant .
Serum ALP levels in estrogen group , genistein group , treadmill group , vibration group and lithium chloride group were significantly lower than that in ovariectomized group .
( 4 ) The bone mineral density , bone volume fraction and trabecular number were significantly higher than those in the ovariectomized group , but there were no significant changes in the bone density , bone volume fraction and the structure model index in the estrogen group , the vibration group and the lithium chloride group , and the expression level of P - gsk - 3.beta . in the estrogen group , the vibration group and the lithium chloride group was significantly higher than that in the ovariectomized group .
There was no significant difference in the expression level of P - gsk - 3 in the femoral and bone marrow cells of genistein group than in the ovariectomized group .
There was a significant difference in the expression levels of the femur , bone marrow cells and P - gsk - 3 beta in the treadmill group than in the ovariectomized group .
( 6 ) The level of ER - 偽 in estrogen group was significantly higher than that in ovariectomized group , tibia , third lumbar vertebra , bone marrow cell and ER - 偽 expression level was significantly lower than that in ovariectomized group .
The expression level of ER - a in the femur of genistein group was significantly higher than that in ovariectomized group , and the expression level of ER - 偽 in tibia and third lumbar vertebra was significantly lower than that in ovariectomized group , but there was no significant difference between bone marrow cells and ER - 偽 expression level .
The expression level of ER - 偽 in the femur of the lithium chloride group was significantly higher than that in the ovariectomized group , and the expression level of ER - 偽 in the tibia , the third lumbar vertebra and the uterus was significantly lower than that in the ovariectomized group , but there was no significant difference in the ER - 偽 expression level of the bone marrow cells .
The expression level of ER - 偽 in the femur , the third lumbar vertebra and the bone marrow cells was significantly higher than that in the ovariectomized group , while the expression level of ER - 偽 in the tibia and uterus was significantly lower than that in the ovariectomized group .
( 7 ) The levels of ER - a protein in the femur , tibia and uterus of the estrogen group were significantly lower than that in the ovariectomized group , while the expression level of the third lumbar vertebra and the bone marrow cell ERR - a protein was significantly higher than that in the ovariectomized group .
The expression of ERR - a protein in femur and tibia of genistein group was significantly lower than that in ovariectomized group , third lumbar vertebra , bone marrow cell and uterus ERR - a protein expression level was significantly higher than that in ovariectomized group .
The expression level of ERR - a protein in the femur , tibia and third lumbar vertebra of the lithium chloride group was significantly lower than that in the ovariectomized group , and the expression level of the ERR - a in the bone marrow cells was significantly higher than that in the ovariectomized group , while the expression level of the ERR - a in the uterus was not significant ;
The expression level of ERR - a protein in the femur , tibia and uterus of treadmill group was significantly lower than that in ovariectomized group , and the expression level of ERR - a protein in the third and third lumbar and bone marrow cells was significantly higher than that in ovariectomized group .
The serum ALP level of ovariectomized osteoporotic rats can be reduced by treadmill movement , vibration , genistein , lithium chloride and estrogen ;
The treadmill exercise can slow down the bone mineral density in ovariectomized osteoporotic rats , while lithium chloride can improve the bone microstructure of ovariectomized osteoporotic rats : treadmill exercise , vibration , genistein , lithium chloride and estrogen are all located and tissue - dependent to regulate the protein expression levels of the ovariectomized osteoporotic rats .
The treadmill exercise , vibration , genistein and lithium chloride have no estrogen - like increase in the side effects of the uterus weight of the ovariectomized osteoporotic rats .
