辛伐他汀與阿侖膦酸鈉對去卵巢大鼠骨丟失及力學性能退變干預效果的比較

【摘要】:目的阿侖磷酸鈉是目前一線抗骨質(zhì)疏松藥物,辛伐他汀作為降脂類藥物因表現(xiàn)出促骨形成作用潛能而備受關注,文中旨在比較辛伐他汀與阿侖膦酸鈉對卵巢切除大鼠骨丟失的作用。方法 6月齡SD大鼠32只,隨機數(shù)字表法分為假手術組、模型組、辛伐他汀組和阿侖膦酸鈉組,每組8只。假手術組:只暴露卵巢,不予切除,直接縫合;其余大鼠接收雙側卵巢切除術。模型組術后給予雙蒸水;辛伐他汀組:10 mg/m L辛伐他汀溶于雙蒸水制成混懸液,給藥劑量5 mg/(kg·d)灌胃;阿侖膦酸鈉組:25μg/m L阿侖膦酸鈉溶液,皮下注射,每周70μg/kg。以上干預措施持續(xù)12周,3個月后處死大鼠。ELISA法分析血清Ⅰ型前膠原氨基端前肽(procollagen typeⅠN-terminal propeptide,PINP)、Ⅰ型膠原羧基端交聯(lián)端肽(cross-linked Ctelopeptides of Type I collagen,ICTP)含量,雙能X線法檢測大鼠第4腰椎骨密度、骨組織形態(tài)計量學分析其松質(zhì)骨骨量和微觀結構,取第5腰椎行壓縮試驗,分析其最大載荷、彈性模量等生物力學性能。結果血清學指標:假手術組PINP、ICTP血清水平均顯著低于其余3組(P0.05),阿侖膦酸鈉組ICTP水平低于模型組和辛伐他汀組(P0.05)。骨密度:模型組骨密度[(0.238±0.007)g/cm~2]低于假手術組[(0.276±0.015)g/cm~2]、辛伐他汀組[(0.250±0.014)g/cm~2]、阿侖膦酸鈉組[(0.269±0.014)g/cm~2],差異均有統(tǒng)計學意義(P0.05),辛伐他汀組顯著低于假手術和阿侖膦酸鈉組(P0.05),假手術組與阿侖膦酸鈉組比較的差異無統(tǒng)計學意義(P0.05)。骨組織形態(tài)計量學分析:模型組骨小梁相對體積[(19.90±1.69)%]顯著低于假手術組[(29.03±2.59)%]和阿侖膦酸鈉組[(27.05±1.91)%],差異有統(tǒng)計學意義(P0.05),骨小梁分離度[(357.33±26.55)μm]顯著高于假手術組[(211.17±16.56)μm]和阿侖膦酸鈉組[(252.50±19.35)μm],差異有統(tǒng)計學意義(P0.05)。生物力學性能:模型組最大載荷和彈性模量均顯著低于其余各組(P0.05)。結論辛伐他汀和阿侖膦酸鈉均可抑制卵巢切除大鼠骨量丟失,兩者對該模型生物力學性能的維持作用無顯著區(qū)別,但后者維持骨密度的作用更加顯著。
[Abstract]:Objective Alendronate is a first-line anti-osteoporosis drug. Simvastatin as a lipid lowering drug has attracted much attention because of its potential to promote bone formation. The purpose of this study was to compare the effects of simvastatin and alendronate on bone loss in ovariectomized rats. Methods Thirty-two 6-month-old SD rats were randomly divided into sham-operated group, model group, simvastatin group and alendronate group with 8 rats in each group. Sham operation group: only exposed ovary, no resection, direct suture; the other rats received bilateral ovariectomy. The model group was given double-steamed water after operation, simvastatin group: 10 mg/m / L simvastatin solution dissolved in double-steamed water to make suspension (5 mg/(kg d), alendronate group: 25 渭 g / mL alendronate sodium solution, subcutaneously injected, 70 渭 g / kg / week.
