【摘要】：目的:通過體外分離培養(yǎng)脂肪干細(xì)胞,并進(jìn)行傳代、表型鑒定及多向誘導(dǎo)分化,進(jìn)一步了解其生物學(xué)特性,為臨床運(yùn)用奠定實(shí)驗(yàn)基礎(chǔ)。方法:取SD大鼠附睪近心端,腎周及腹股溝韌帶皮下脂肪組織,采用密度梯度離心和貼壁培養(yǎng)相結(jié)合方法,對(duì)脂肪干細(xì)胞進(jìn)行分離、體外培養(yǎng)及擴(kuò)增、表型鑒定及成骨成脂誘導(dǎo)分化,在倒置顯微鏡下觀察原代及傳代細(xì)胞形態(tài)變化,并采用MTT法檢測(cè)第3、5、7、9代細(xì)胞增殖能力,用流式細(xì)胞儀檢測(cè)細(xì)胞表面抗原標(biāo)記,取第3代細(xì)胞進(jìn)行成骨成脂誘導(dǎo)分化,并采用茜素紅染色、油紅O染色方法進(jìn)行鑒定。結(jié)果:采用密度梯度離心和貼壁培養(yǎng)相結(jié)合方法得到的脂肪干細(xì)胞,其細(xì)胞形態(tài)較為均一,基本呈長梭形,旋渦狀生長,并形成集落樣生長。MTT法檢測(cè)第3、5、7、9代ADSCs呈典型的"S"形增殖曲線,符合干細(xì)胞的特性,脂肪干細(xì)胞表型鑒定結(jié)果顯示CD29陽性高表達(dá),CD34陰性表達(dá),CD45低表達(dá),CD105弱陽性表達(dá),脂肪干細(xì)胞能向成骨細(xì)胞及脂肪細(xì)胞分化。結(jié)論:脂肪干細(xì)胞取材較方便,含量豐富、并能夠在體外分離培養(yǎng),其穩(wěn)定表達(dá)特異性的表面抗原,并且經(jīng)過相應(yīng)誘導(dǎo)培養(yǎng)后可向成骨及成脂多向分化,可作為優(yōu)良的骨組織工程的種子細(xì)胞來源。
[Abstract]:Objective: to investigate the biological characteristics of adipose stem cells (ASCs) and to establish the experimental basis for clinical application by means of isolation and culture in vitro, passage, phenotypic identification and multidirectional differentiation. Methods: the adipose tissues of the proximal epididymis, perirenal and inguinal ligaments of SD rats were isolated, cultured and expanded in vitro by density gradient centrifugation and adherent culture. Phenotypic identification and osteogenic adipogenic differentiation were used to observe the morphological changes of primary and passage cells under inverted microscope. MTT assay was used to detect the proliferative ability of passage 3 ~ (5) and 7 ~ (th) passage cells, and flow cytometry was used to detect cell surface antigen labeling. Osteogenic adipogenic differentiation of the third passage cells was carried out and identified by alizarin red staining and oil red O staining. Results: the adipose stem cells were obtained by density gradient centrifugation and adherent culture. The results showed that the proliferation curve of ADSCs in the 3rd generation was "S" shape, which was in line with the characteristics of stem cells. The phenotypic identification of adipose stem cells showed that the positive expression of CD29 was high, the negative expression of CD34 was negative, the expression of CD45 was low and the expression of CD105 was weak. Adipose stem cells can differentiate into osteoblasts and adipocytes. Conclusion: adipose stem cells are convenient and rich in content, and can be isolated and cultured in vitro. They can express specific surface antigen stably, and can differentiate into osteogenesis and adipogenesis after induction and culture. It can be used as a good seed cell source for bone tissue engineering.
