Tim-1蛋白表達(dá)及其基因多態(tài)性與系統(tǒng)性紅斑狼瘡患者營(yíng)養(yǎng)功能的相關(guān)性

【摘要】:目的探討Tim-1蛋白表達(dá)及其基因多態(tài)性與系統(tǒng)性紅斑狼瘡(SLE)患者營(yíng)養(yǎng)功能指標(biāo)的相關(guān)性。方法收集126例SLE患者外周血樣;采用ELISA法檢測(cè)血清中人Tim-1蛋白水平;采用PCR-RFLP法檢測(cè)外周血DNA樣本Tim-1基因-416GC和-1454GA多態(tài)性;應(yīng)用免疫比濁法檢測(cè)血清標(biāo)本中前白蛋白、銅藍(lán)蛋白、視黃醇結(jié)合蛋白的水平;采用電化學(xué)發(fā)光法檢測(cè)血清標(biāo)本中鐵蛋白、1,25-二羥維生素D3的水平。結(jié)果血清Tim-1蛋白、前白蛋白、銅藍(lán)蛋白、視黃醇結(jié)合蛋白、鐵蛋白及1,25-二羥維生素D3的濃度分別為(249.7±30.2)pg/m L、(226±42)μg/m L、(363±95)μg/m L、(29.4±13.2)μg/m L、(355±164)ng/m L、(26.4±11.5)ng/m L;-416GC位點(diǎn)上GG基因型占11.9%,GC基因型占57.1%,CC基因型占31.0%;-1454GA位點(diǎn)上GG基因型占67.5%,GA基因型占26.2%,AA基因型占6.3%。-416GC位點(diǎn)不同基因型之間(F=0.575,P=0.564)及-1454GA位點(diǎn)不同基因型之間(F=1.255,P=0.289)Tim-1蛋白濃度均無(wú)統(tǒng)計(jì)學(xué)差異。Tim-1與前白蛋白顯著負(fù)相關(guān)(r=-0.176,P=0.033),與銅藍(lán)蛋白(r=0.205,P=0.014)和1,25-二羥維生素D3(r=0.166,P=0.042)均顯著正相關(guān)。-1454GA位點(diǎn)GG基因型患者血清中前白蛋白水平顯著降低(P=0.027),AA基因型患者血清中1,25-二羥維生素D3水平顯著降低(P=0.024)。結(jié)論血清Tim-1蛋白水平及Tim-1基因-1454GA位點(diǎn)多態(tài)性與SLE患者營(yíng)養(yǎng)功能指標(biāo)存在相關(guān)性。
[Abstract]:Objective to investigate the relationship between Tim-1 protein expression and gene polymorphism in patients with systemic lupus erythematosus (SLE). Methods Peripheral blood samples were collected from 126 patients with SLE, serum Tim-1 protein levels were detected by ELISA method, Tim-1 gene -416GC and -1454GA polymorphisms in peripheral blood DNA samples were detected by PCR-RFLP method, prealbumin and ceruloplasmin in serum samples were detected by immunoturbidimetric method. The level of retinol binding protein and the level of ferritin 1 / 25-dihydroxyvitamin D _ 3 in serum samples were determined by electrochemiluminescence (ECL). Results Serum Tim-1 protein, prealbumin, ceruloplasmin, retinol binding protein, The concentrations of ferritin and 1-dihydroxyvitamin D _ 3 were 249.7 鹵30.2)pg/m / L ~ (226 鹵42) 渭 g / mL ~ (63 鹵95) 渭 g / m ~ (-1)) 29.4 鹵13.2) 渭 g / m ~ (1 / 3) ~ (355) 鹵164)ng/m ~ (26.4) 鹵11.5)ng/m / L ~ (26.4) 鹵11.5)ng/m / L ~ (416GC), respectively. There was no significant difference in the protein concentration between the two genotypes. Tim-1 was negatively correlated with prealbumin, and was significantly negatively correlated with prealbumin, and positively correlated with r0.205P0.014) and 125-dihydroxyvitamin D3HV 0.166P0.042) in the serum of GG genotype patients with GG locus 1454GA. The level of albumin in serum of patients with AA genotype was significantly lower than that of control group (P < 0. 027), and the serum level of vitamin D 3 was significantly lower than that of control group (P < 0. 024). Conclusion the level of serum Tim-1 protein and the polymorphism of Tim-1 gene -1454GA are correlated with the nutritional function of SLE patients.
