磁標(biāo)記內(nèi)皮祖細(xì)胞移植對糖尿病大鼠腎功能改善的作用:MRI成像驗證

【摘要】:背景:近幾年的諸多研究發(fā)現(xiàn),內(nèi)皮祖細(xì)胞在損傷性疾病等方面有重要作用。目的:探討磁標(biāo)記內(nèi)皮祖細(xì)胞移植對糖尿病大鼠腎功能的改善作用及MRI成像。方法:將60只Wistar大鼠隨機(jī)分為3組,正常對照組不作任何處理;模型組、實驗組腹腔注射40 mg/kg鏈脲佐菌素,建立1型糖尿病模型,造模后4周,實驗組尾靜脈注射磁標(biāo)記內(nèi)皮祖細(xì)胞懸液0.15 m L(細(xì)胞濃度1×109 L-1)。細(xì)胞移植8周后,檢測3組大鼠空腹血糖、血清胰島素、血肌酐和尿素氮及24 h尿蛋白水平;采用MRI對移植細(xì)胞進(jìn)行活體示蹤,并與腎臟組織切片進(jìn)行對照;測量大鼠體質(zhì)量及腎質(zhì)量變化,計算腎質(zhì)量指數(shù)。結(jié)果與結(jié)論:(1)與正常對照組比較,模型組空腹血糖、血肌酐和尿素氮、24 h尿蛋白及腎質(zhì)量指數(shù)升高(P0.05),胰島素水平降低(P0.05);(2)與模型組比較,實驗組空腹血糖、血肌酐、尿素氮、24 h尿蛋白及腎質(zhì)量指數(shù)降低(P0.05),胰島素水平升高(P0.05);(3)實驗組可見稍長T1短T2信號,病灶邊緣部位明顯,梯度回波FLASH序列較T2加權(quán)RARE序列明顯,其余組均未見明顯低信號改變;實驗組可見磁標(biāo)記的陽性細(xì)胞且與組織切片結(jié)果相吻合,模型組和正常對照組未見磁標(biāo)記的陽性細(xì)胞;(4)結(jié)果表明,磁標(biāo)記內(nèi)皮祖細(xì)胞移植可在一定程度上改善糖尿病大鼠的腎功能紊亂。
[Abstract]:Background: in recent years, many studies have found that endothelial progenitor cells play an important role in injurious diseases.Objective: to investigate the effect of magnetically labeled endothelial progenitor cell transplantation on renal function and MRI imaging in diabetic rats.Methods: sixty Wistar rats were randomly divided into 3 groups without any treatment in the normal control group, and in the experimental group, 40 mg/kg streptozotocin was injected intraperitoneally to establish the model of type 1 diabetes.The experimental group was injected with magnetic labeled endothelial progenitor cell suspension of 0.15 mL (cell concentration 1 脳 10 9 L -1).After 8 weeks of cell transplantation, fasting blood glucose, serum insulin, serum creatinine, urea nitrogen and 24 h urine protein levels were measured in three groups, and MRI was used to trace the transplanted cells in vivo and compared with renal tissue sections.The body mass and renal mass of rats were measured and the renal mass index was calculated.Results compared with the normal control group, the fasting blood glucose, serum creatinine and urea nitrogen nitrogen 24 h urinary protein and renal mass index in the model group were increased (P 0.05), the insulin level was decreased (P 0.05) and compared with the model group, the fasting blood glucose and serum creatinine in the model group were significantly higher than those in the control group.Urinary protein and renal mass index (RMI) were decreased in 24 h in urea nitrogen group (P 0.05) and insulin level was increased (P 0.05). In the experimental group, a little longer T 1, short T 2 signal was observed, the edge of the lesion was obvious, and the gradient echo FLASH sequence was more obvious than that of T 2 weighted RARE sequence.There were no significant changes in low signal intensity in the other groups, and the positive cells were observed in the experimental group, which were consistent with the results of tissue sections, but not in the model group and the normal control group (P < 0.05).Magnetic labeled endothelial progenitor cell transplantation can improve renal dysfunction in diabetic rats to some extent.
