高效液相色譜-電化學檢測法研究大蒜素對百草枯中毒小鼠腦組織中單胺類神經(jīng)遞質含量的影響

【摘要】:建立了單胺類神經(jīng)遞質的高效液相色譜-電化學檢測方法,探討了大蒜素對百草枯中毒小鼠腦組織內的單胺類神經(jīng)遞質及其代謝產(chǎn)物的影響。健康ICR小鼠隨機分為4組:正常對照(Con)組、大蒜素(DAS)組、百草枯染毒(PQ)組和大蒜素治療(PQ+DAS)組,每組6只。PQ組灌胃PQ 35 mg/kg建立中毒模型,Con組和DAS組灌服同體積生理鹽水。DAS組和PQ+DAS組小鼠于造模2、26、50 h后腹腔內各注射1次DAS(25 mg/kg),PQ組和正常組小鼠腹腔內注射等量無菌生理鹽水。造模12 h后處死小鼠。收集小鼠腦組織樣品并稱重,處理后進樣。色譜分析采用Acclaim 120 C18(150 mm×2.1 mm,3μm)色譜柱,以離子對緩沖鹽溶液-甲醇(93∶7,體積比)為流動相,流速為0.27 m L/min;電化學檢測器檢測,工作電壓為450 m V。結果表明,與正常組和DAS組比較,PQ組部分神經(jīng)遞質水平降低;DAS治療后能夠改變上述生化指標的變化;與PQ組相比,PQ+DAS組神經(jīng)遞質含量增高,差異均具有統(tǒng)計學意義(P0.05)。腦組織樣品中5-HT、DA、5-HIAA、DOPAC和HVA低、中、高3水平的加標回收率均為79.8%~112.1%,相對標準偏差(RSD)小于10%。實驗表明,大蒜素對百草枯中毒后小鼠體內含量降低的神經(jīng)遞質有明顯的上調作用。
[Abstract]:A high performance liquid chromatography-electrochemical detection method for monoamine neurotransmitters was established. The effects of allicin on monoamine neurotransmitters and their metabolites in the brain of paraquat poisoned mice were investigated. Healthy ICR mice were randomly divided into 4 groups: normal control group (Cona), allicin group (Das). Paraquat (PQQ) group and allicin treatment group (PQ DAS) group, Six mice in each group were given PQ35 mg/kg by intragastric administration of PQ35 mg/kg in each group. The mice in the Con group and the DAS group were given the same volume of normal saline. DAS and PQ DAS for 50 hours after the model was made. There was no equal amount of intraperitoneal injection of DAS(25 mg / kg PQ and normal mice in the control group and once a DAS(25 mg / kg group. The mice were killed after 12 hours of modeling. The brain tissue samples were collected and weighed. After treatment, the chromatographic analysis was performed on Acclaim 120C18C18mm 脳 2.1mm ~ (3 渭 m) column. The mobile phase consisted of ion-pair buffer salt solution and methanol (93: 7, volume ratio) at a flow rate of 0.27 mL / min. The working voltage was 450 MV by electrochemical detector. Compared with the normal group and the DAS group, the level of some neurotransmitters in the PQ group decreased and the changes of the above biochemical indexes were changed after treatment, and the neurotransmitter content in the PQ DAS group was higher than that in the PQ group, and that in the PQ DAS group was higher than that in the PQ group. The difference was statistically significant (P 0.05). The recoveries of 5-HTDDAN 5-HIAADOPAC and HVA in brain tissue samples were all 79.8% and 112.1, respectively, and the relative standard deviation (RSD) was less than 10%. Allicin significantly up-regulated the neurotransmitters of paraquat poisoning in mice.
