高原紅細胞增多癥血漿差異蛋白組學的初步研究

【摘要】:目的:應用串聯(lián)質譜標簽(TMT)結合質譜技術比較高原紅細胞增多癥(HAPC)患者和健康對照者血漿中的差異表達蛋白。方法:收集HAPC患者血漿4例,與之相匹配的健康對照組血漿5例,組內等量混合后利用多重免疫親和層析柱去除14種高豐度蛋白,TMT試劑標記后樣本進行液相色譜分離,采用HPLC-MS/MS質譜鑒定及相對定量;HAPC患者和健康對照各20例血漿,ELISA方法驗證篩選的差異蛋白。結果:共鑒定和定量了1094種蛋白,其中差異表達的蛋白有249種,其中HAPC組較健康對照組表達量上調≥1.5倍的蛋白質有162種,表達量下調≤0.67倍的蛋白質有87種,炎性相炎癥相關差異表達蛋白17種;炎癥蛋白CRP和SAA在HAPC組中較健康對照組表達量均上調(P0.05)。結論:篩選出17種與炎癥相關的差異表達蛋白,為深入研究HAPC發(fā)生機制奠定了良好基礎。
[Abstract]:Objective: to compare the differentially expressed proteins in plasma of patients with high altitude polycythemia (HAP) and healthy controls by tandem mass spectrometry (tandem mass spectrometry) and mass spectrometry. Methods: plasma samples from 4 patients with HAPC and 5 healthy controls were collected. The samples were separated by liquid chromatography after being mixed in the same amount within the group, 14 kinds of high abundance protein TMT reagent were removed by multiple immunoaffinity chromatography column, and the samples were labeled with TMT reagent. The differential proteins were identified by HPLC-MS/MS mass spectrometry and 20 patients and 20 healthy controls by Elisa respectively. Results: 1094 proteins were identified and quantified, of which 249 were differentially expressed.
