表沒食子兒茶素沒食子酸酯對高尿酸血癥大鼠血管內(nèi)皮炎癥因子的作用

【摘要】:目的探討表沒食子兒茶素沒食子酸酯(EGCG)對高尿酸血癥大鼠血管內(nèi)皮炎癥因子的作用。方法將36只雄性SD大鼠分為正常對照組、模型對照組、EGCG低劑量組、EGCG中劑量組、EGCG高劑量組、別嘌呤醇組,5周后,運用RT-PCR檢測大鼠主動脈MCP-1、TNF-α、ICAM-1、ET-1及e NOS表達量,以及大鼠血清尿酸、肌酐、尿素氮和黃嘌呤氧化酶(XO)水平。結(jié)果模型對照組大鼠的血尿酸和XO水平顯著高于正常對照組(P0.01),EGCG各濃度組及別嘌呤醇組大鼠的血尿酸和XO水平與模型對照組相比較則有不同程度下降(P0.01)。模型對照組大鼠主動脈MCP-1、TNF-α、ICAM-1的mRNA表達量較正常對照組顯著升高(P0.01),EGCG各濃度組和別嘌呤組大鼠主動脈TNF-α、ICAM-1的mRNA表達量與模型對照組相比顯著下降(P0.01),EGCG高劑量組和別嘌呤組MCP-1 mRNA表達量與模型對照組相比顯著下降(P0.05)。EGCG中劑量組和別嘌呤組ET-1 mRNA表達量與模型對照組相比顯著下降(P0.05)。結(jié)論 EGCG干預(yù)可以降低高尿酸血癥大鼠的血尿酸水平,同時減少血管內(nèi)皮炎癥因子的表達。
[Abstract]:Objective to investigate the effect of epigallocatechin gallate (EGCG) on vascular endothelial inflammation in hyperuricemia rats. Methods 36 male SD rats were divided into normal control group and model control group. After 5 weeks in the allopurinol group, the expression of MCP-1TNF- 偽 ICAM-1 ET-1 and e NOS, serum uric acid and creatinine in rat aorta were detected by RT-PCR. Results the levels of serum uric acid and XO in the model control group were significantly higher than those in the normal control group (P 0.01) and the allopurinol group and the rats in the allopurinol group were higher than those in the model control group. The expression of ICAM-1 mRNA in aorta of the model control group was significantly higher than that of the normal control group. The mRNA expression of TNF- 偽 ICAM-1 in the aorta of the model control group was significantly lower than that of the model control group. The expression of MCP-1 mRNA in the dose group and allopurine group was significantly decreased compared with the model control group. The expression of ET-1 mRNA in the middle dose group and allopurine group was significantly lower than that in the model control group. Conclusion EGCG intervention can reduce the level of serum uric acid in hyperuricemia rats. At the same time, the expression of vascular endothelial inflammatory factor was reduced.
