C1型尼曼-匹克病小鼠生長繁殖及血液生理生化指標(biāo)的測定分析

【摘要】:目的測定SPF級C1型尼曼-匹克病小鼠(Npc1~(-/-)小鼠)的生長繁殖及血液生理生化指標(biāo),為開展NPC1病人的研究提供理論性的基礎(chǔ)資料。方法 (1)選取0~77日齡Npc1~(-/-)、Npc1~(+/-)和Npc1+/+小鼠雌雄各40只,定期稱重并繪制生長曲線;(2)Npc1~(-/-)小鼠由Npc1~(+/-)小鼠交配繁殖產(chǎn)生,統(tǒng)計Npc1~(+/-)小鼠連續(xù)4代的繁殖數(shù)據(jù);(3)檢測60日齡Npc1~(-/-)和Npc1+/+小鼠的血液生理生化指標(biāo)。結(jié)果 (1)Npc1~(-/-)小鼠的體重在7周前隨著日齡的增加而增加,7周后隨著日齡的增加而減少,直至11周左右死亡。Npc1+/+和Npc1~(+/-)小鼠的體重均隨著日齡的增長而逐漸增加,兩者之間并沒有顯著差別。4周后雄性比雌性體重增加明顯比雌性小鼠快;(2)Npc1~(+/-)小鼠不同代內(nèi)交配分娩間隔、窩產(chǎn)仔數(shù)、離乳仔數(shù)、離乳仔數(shù)中雌雄數(shù)量及陽性數(shù)量差異無顯著性(P0.05),而第2代的離乳率明顯大于第1代(P0.05);(3)Npc1~(-/-)和Npc1+/+小鼠血液生理指標(biāo)中平均紅細(xì)胞血紅蛋白含量(MCH)和平均過氧化物酶指數(shù)(MPXI)差異有顯著性(P0.05),其余差異無顯著性(P0.05)。生化指標(biāo)中尿素(UREA)差異有極顯著性(P0.01),而天門冬氨酸氨基轉(zhuǎn)移酶(AST)、葡萄糖(GLU)、乳酸脫氫酶(LDH)、鉀(K)和銅(Cu)等差異有顯著性(P0.05),其余差異無顯著性(P0.05)。結(jié)論 (1)Npc1~(-/-)、Npc1~(+/-)和Npc1+/+小鼠的生長曲線因基因型和性別的不同而存在差異;(2)Npc1~(+/-)小鼠不同代的繁殖能力差異無顯著性;(3)Npc1~(-/-)和Npc1+/+小鼠的部分血液生理生化指標(biāo)差異有顯著性。
[Abstract]:Objective to determine the growth, reproduction and blood physiological and biochemical indexes of SPF grade C1 Nieman-Pick disease mice) and to provide theoretical data for the study of NPC1 patients. Weighing and drawing the growth curve on a regular basis, the Npc1 + / -r-) mice are produced by mating and reproduction in the Npc1- (/ -) mice. The blood physiological and biochemical indexes of 60 day old Npc1 +% -r-) and Npc1 / mice were measured. Results the body weight of Npc1 / Npc1 + /-/ -) mice increased 7 weeks ago with the increase of age, and then decreased with the age of 7 weeks. The body weight of both Npc1 / Npc1 / Npc1 / and Npc1 (/ -) mice gradually increased with the age of 11 weeks or so. There was no significant difference between the two groups. After 4 weeks, the male weight increased significantly faster than that of the female mice, and the mating interval, litter size and litter size of Npc1 (/ -) mice in different generations were significantly faster than that in female mice. There was no significant difference in the number of female and male and the positive number of female and male in litter size, but the rate of weaning in the second generation was significantly higher than that in the first generation (P 0.05) and the mean RBC hemoglobin content and peroxidase finger in the blood physiological indexes of Npc1 / mice. The difference of MPXI was significant (P0.05), but the other differences were not significant (P0.05). There was a very significant difference in Urea (Urea) among biochemical indexes, but there was a significant difference in aspartate aminotransferase (ASTT), glucose gluconium (GLU), lactate dehydrogenase (LDH), potassium (K) and copper (Cu2). Conclusion there is no significant difference in growth curves between NPC1 (P / P) and Npc1 / mice according to genotype and sex. There is no significant difference in reproductive ability of different generations of Npc1C ~ (-1) (P ~ -) and Npc1 / Npc1 / mice. There is no significant difference in the reproductive ability of different generations of mice and some of the blood physiological growth of Npc1 / mice (P ~ (-1)) and Npc1 / mice (P ~ (-1) -P ~ (-1)) and / or Npc1 / mice (P ~ (-1) -P ~ (-)) and Npc1 / Npc1 / mice, there is no significant difference in the reproductive ability of different generations of mice. There was significant difference in chemical index.
